The radiosensitizing potential of DNA-PK inhibitors in breast cancer targeted radiotherapy (EANM 2026) - Sep 29, 2026 - Abstract #EPS-019; Pres time: Oct 18, 2026; 08:00 AM - 09:30 AM; Location: Hall G2; "Materials and We analyzed the viability of two breast carcinoma cell lines, MDA-MB-231 and MCF-7, in the presence of three DNA-PK inhibitors: CC-115, M3814 and PIK-75 HCl, selected based on in-silico binding studies, as well as their difference in affinities and mechanism of action. The cells were exposed to four concentrations of the inhibitors, Doxorubicin as the main DSB inducer, to gain an overview of the pathway... The implementation of DNA-PK inhibitors into medical practice can enable the stratification of oncologic patients into two categories, based on the tumors' vulnerability to NHEJ disruptions. Thus, the therapeutic pathways of patients with NHEJ tumors could branch, combining traditional genotoxic therapies (radiation and DNA-damaging chemotherapeutics) with DNA-PK inhibitors to achieve an enhanced effect and improved survival outcomes."
