Janus Kinase Inhibitors Rapidly Reduce T-Cell Immunosenescence and Epigenetic Age Acceleration in Rheumatoid Arthritis (ACR Convergence 2026) - Sep 8, 2026 - Abstract #84; Pres time: Nov 8, 2026; 10:30 AM - 12:30 PM; Location: WB1&2; In Person; "We assessed whether Janus kinase inhibitors (JAKi) can modify senescent/exhausted T-cell phenotypes and residual epigenetic age acceleration in active RA Forty-eight patients with active RA starting a JAKi (tofacitinib, baricitinib, upadacitinib or filgotinib) were evaluated at baseline and after 3 months of treatment; 31 age- and sex-matched healthy controls (HCs) were included. JAKi therapy induced a partial reversal of T-cell immunosenescence and a robust reduction in integrated DNAm age acceleration in RA. These data support immune ageing as a therapeutically modifiable dimension of RA biology and suggest that JAK/STAT inhibition may rapidly remodel ageing-related immune and epigenetic phenotypes. JAKi therapy induced a partial reversal of T-cell immunosenescence and a robust reduction in integrated DNAm age acceleration in RA."
