The Selective PI3KδInhibitor Roginolisib Synergizes With JAK Inhibitors in Progenitor Cells From Naïve and JAK Inhibitor–Refractory/Resistant Patients With Myelofibrosis (SOHO 2026) - Sep 1, 2026 - Abstract #MPN-545; Location: LEVEL 3, HALL B3; P1/2 | " Roginolisib alone did not inhibit proliferation in JAK2 V617F UKE-1 cells up to 10 μM, whereas ruxolitinib (Ruxo) showed the expected activity...Combinations with Ruxo or momelotinib led to marked, synergistic inhibition of colony formation (CI, 0.07–0.5), reaching ∼70% to 80% suppression... Roginolisib shows activity in primary MF cells and demonstrates strong synergy with JAK inhibitors. These results support the ongoing phase 1b trial of roginolisib in MF patients with suboptimal responses to JAKi (HEMAMED, NCT06887803).
