From Genetic Risk to Tissue Circuitry: IL-23-Driven Remodeling of Stromal-Immune Niches in Psoriasis (ESDR 2026) - Sep 9, 2026 - Abstract #147; Pres time: Sep 9, 2026; 10:00 AM - 04:00 PM; Location: Hall 2B; "To address this, single-cell RNA sequencing, TCR sequencing, and spatial transcriptomics were performed on matched pre- and post-treatment psoriatic skin samples from patients (n=16) treated with the anti–IL-23 monoclonal antibody risankizumab (300 or 600 mg at Weeks 0, 4, and 16), followed without further dosing until Week 52...Notably, both fibroblast-derived and vascular niche signals were attenuated following IL-23 blockade. Together, these findings demonstrate how genetic risk converging on IL-23 signaling is translated into pathogenic cellular circuits, and how targeted blockade of this pathway disrupts stromal–immune interactions and sustains long-term therapeutic responses."
