AMPA receptor potentiation remains a promising strategy, but future candidates will need to combine potency, finely tuned efficacy, favorable pharmacokinetics, and circuit-level selectivity. The most successful next-generation PAMs will likely be those that maximize therapeutic window while minimizing seizure liability."
