Deciphering DNA Repair Mechanisms Across Targeted Radionuclide Therapy and Other Radiotherapy Modalities via a CRISPR/Cas9 DNA Repair Gene Knockout Screen (EANM 2026) - Sep 29, 2026 - Abstract #EP-0083; "Aim/Introduction: Targeted radionuclide therapy (TRT), exemplified by [177Lu]Lu-PSMA-617 (Pluvicto®) has become an effective treatment option for castration resistant metastatic prostate cancer, providing hope for patients beyond conventional options... These results underscore the importance of directly comparing radiation modalities to define modalityspecific sensitivities and DNA damage-repair pathway dependencies, which are critical for optimizing treatment and guiding rational combination strategies. Future work will extend this approach by engineering RPE1 cells to express prostatespecific membrane antigen (PSMA), enabling direct comparison of nontargeted and PSMAtargeted treatments using [¹⁷⁷Lu] Lu- and [²²⁵Ac]AcPSMA617. In parallel, CRISPRKO screens in PSMApositive LNCaP prostate cancer cells will aim to uncover cancerspecific DNA repair vulnerabilities with potential clinical relevance."
