Transketolase Inhibition Sensitizes PSMA-Targeted 177 Lu Radioligand Therapy in PSMA-Positive Tumor Models (EANM 2026) - Sep 29, 2026 - Abstract #OP-480; Pres time: Oct 19, 2026; 04:45 PM - 06:15 PM; Location: Hall E2; "Transketolase inhibition with B-OT metabolically sensitizes PSMA-positive prostate tumors to 177Lu-PSMA-617 without impairing tumor uptake or pharmacokinetics. Metabolomic analyses show that B-OT disrupts nucleotide and energy metabolic homeostasis, thereby creating a metabolically vulnerable state that limits tumor adaptation to radioligand-induced stress. Quantitative imaging and therapeutic data support metabolic radiosensitization as a promising strategy to overcome resistance in PSMA-targeted radioligand therapy, with mechanistic validation ongoing."
