Comparison of Ciprofol Versus Propofol for the General Anaesthesia During Gynecological Day Surgery: A Prospective, Randomized, Double-Blind, Non-Inferiority Trial.

- "Anesthesia induction in the ciprofol group: ciprofol 0.5 mg/kg was given intravenously within 1 minute, alfentanil 20 μg/kg was given intravenously within 30 seconds, mivacurium 0.2 mg/kg was given intravenously within 30 seconds, and anesthesia maintenance: ciprofol 1.25 mg/kg/h combined with alfentanil 40 μg/kg/h was given intravenously. However, the awakening time of the ciprofol group was significantly longer than that of the propofol group (7.57 vs 5.52 min). Ciprofol demonstrated non-inferior efficacy to propofol for gynecological day surgery anesthesia, while offering superior hemodynamic stability and reduced adverse effects."

- "For elderly male patients undergoing transperineal prostate biopsy under intravenous anesthesia with preservation of spontaneous breathing, the combined use of remimazolam and alfentanil can achieve more stable intraoperative hemodynamics, faster recovery from anesthesia and a lower incidence of perioperative adverse reactions, and produce an even better anesthetic effect with a small dose of propofol to suppress the body movement at the time of needle puncture."

- "These patients were treated with medically approved novel synthetic opioids (fentanyl, remifentanil, sufentanil, or alfentanil) and traditional opioids (morphine, methadone, and hydromorphone) during their clinical treatment. The metabolite ratio of β-hydroxyfentanyl to fentanyl was particularly well suited for children and neonatal patients. In conclusion, concentration ranges, metabolite ratios, and dose correlations of the studied opioids in pediatric hair were established, providing insights into opioid incorporation pathways."

- "Among all interventions, dexmedetomidine combined with alfentanil was the most effective in reducing ED risk for tonsillectomy/adenoidectomy, achieving a SUCRA ranking of 94.63% (logRR = -2.82). Targeted use of dexmedetomidine-based combinations was found to be particularly effective across various surgeries, while fentanyl and propofol-midazolam combinations excelled in specific contexts. This study underscores the importance of tailoring anesthetic adjuvant strategies to specific surgical sites to reduce the risk of ED in pediatric patients, and provides a valuable reference for optimizing anesthetic care in this vulnerable population."

- P=N/A | "For women undergoing hysteroscopic surgery, a dose of 10-12 µg·kg-1 of alfentanil was associated with significant ciprofol-sparing effect, earlier anesthesia emergence, better postoperative analgesia, and less unexpected hemodynamic events compared with sufentanil, but 14 µg·kg-1 alfentanil had the risk of transient desaturation and delayed anesthesia recovery. Indications and the optimal dose of alfentanil in this patient population need further clarification."

Pharmacological interventions for the prevention of pain during endotracheal suctioning in ventilated neonates.

- "Morphine may have little to no effect on PIPP or duration of ventilation (evidence very uncertain). Morphine may not increase the risk of hypotension and probably does not reduce the incidence of severe IVH. Alfentanil may reduce CHEOPS and heart rate change from baseline (evidence very uncertain). Meperidine may reduce BPS. Compared to midazolam, morphine may have little to no effect on PIPP scores, and may result in a large reduction in the duration of ventilation (evidence very uncertain). No studies reported on physiological and behavioural indicators or haemodynamic changes. Morphine may not reduce the incidence of severe IVH, compared to midazolam (low-certainty evidence). Compared to placebo, midazolam may reduce PIPP scores, and has little to no effect on the duration of ventilation or on the risk of severe IVH (low to very low-certainty evidence); ketamine may reduce CHEOPS change, and have little to no effect on heart rate change, or on mean arterial blood..."

Quantitative Prediction of Drug-Drug Interactions Arising from CYP3A4 Induction using Chimeric Mice with Humanized Liver.

- "After repeated administration of rifampicin or efavirenz as CYP inducers to chimeric mice, the relative expression of human CYP3A4 in their livers was measured and plotted against the area under the concentration-time curve (AUC) of rifampicin and efavirenz, respectively, in plasma on the final day of administration. The intrinsic clearance was incorporated into a constructed physiologically based pharmacokinetic model, which successfully predicted the pk change of midazolam or alfentanil coadministered with a CYP inducer or not. The results confirmed that our approach is useful to improve the prediction accuracy of CYP3A4 induction in the preclinical phase."
