CLINICAL PERFORMANCE OF THE NEUMODX HPV ASSAY IN STM MEDIUM IN CERVICAL CANCER SCREENING SETTINGS

- "We assessed the validity of the NeuMoDx for cervical cancer screening purposes, via comparison to a clinically validated comparator assay (digene Hybrid Capture 2; HC2) in cervical specimens collected in digene Specimen Transport Medium (STM). A clinical validation panel comprised of 919 residual STM samples obtained from women aged 21-64 years participating in the organized Slovenian screening program, of these 835 were from women undergoing their regular screening visit after a previous negative screen test while 84 were from women with underlying cervical intraepithelial neoplasia grade 2 or worse (CIN2+) lesions. The clinical sensitivities were 95.2% (80/84; 95%CI 88.0-98.2) and 94.0% (79/84; 95%CI 86.5-97.5) for the NeuMoDx and HC2 assay, respectively, while clinical specificities were 88.9% (742/835; 95%CI 86.8-90.6) and 89.3% (746/835; 95%CI 87.3-91.1) for NeuMoDx and HC2, respectively. The NeuMoDx HPV Assay demonstrated good performance in cervical samples..."

SPRINT-HPV: HR-HPV DETECTION FROM CERVICAL SAMPLES IN POINT OF CARE

- "We developed a POC system that detects HPV types 16, 18, and nine other HR-HPV types. Cervical brush samples were collected at AIIMS in New Delhi, India, and stored using a Digene specimen transport medium (STM)... We developed and performed pilot testing of a portable isothermal POC system for HPV detection from cervical samples in low-resource settings. Further validation of the system is needed to establish analytic performance, especially for non-HPV16/18 genotypes. This fully integrated sample-to-answer platform offers a rapid, cost-effective solution for HR-HPV screening in LMICs."

- "The performance of Roche Cobas 4800 HPV test was validated and compared with Digene Hybrid Capture 2 (HC2) high-risk HPV DNA test for primary screening in a large Chinese screening cohort...The Cobas test demonstrated significantly higher specificity in identifying CIN2+/CIN3+ cases than HC2 test (66.46% vs 43.67% and 65.42% vs 42.86%, p<0.001), with comparable sensitivity in clinical evaluation. Increased specificity of Cobas test would accent women having the highest risk of developing CIN2+, with the potential to reduce unnecessary colposcopy referral in a screening population."

- "Introduction/Background* The aim of this study was to determine the efficiency of the Hybrid Capture 2 (HC2; Digene, Qiagen, Hilden Germany) human papillomavirus (HPV) assay for the detection of cervical neoplasia compared to conventional cytology testing...Conclusion* Implementing a HC2 based cervical cancer screening algorithm could potentially reduce the number of colposcopy/biopsy referrals by over 50% without a substantial impact on the CIN2+ detection rate and no impact on the CIN3+ detection rate. This in turn decreases the associated costs of screening, decreases the number of unnecessary diagnostic procedures and, could increases patient comfort, satisfaction and adherence to the screening program"

In conclusion, HPV infection affects all socioeconomic classes in Brazil, evidencing the importance of offering the HPV vaccine to the entire population."

Self-collection can have a positive impact on a cervical screening program in South Africa by increasing coverage of women in rural areas, in particular those unable to visit the clinics and women attending clinics where cytology-based programs are not functioning effectively."

Performance analysis of high-throughput HPV testing on three automated workflows.

- "During a 3 months period, four technicians were involved in processing 5000 specimens per month on three automated platforms, (1) Qiagen Digene® HC2 HPV DNA test (HC2, signal amplification); (2) Roche Cobas® HPV test (DNA amplification), and (3) Hologic Aptima® HPV test (RNA amplification)...Although all HPV tests used in this comparison meet the international test guidelines, the performance (workflow) characteristics of the assays vary widely. A specific choice of a lab for high throughput testing can be different based on the labs demands, but also hands-on-time, time-to-results / # samples, maintenance, pre-run, run and post-run parameters, consumables, technical support and number of errors are important operational factors for the selection of a fully automated workflow for hrHPV testing."
