Preclinical development of AVB-406: An intravenous AAV-miRNA achieving sustained MAPT knockdown for the treatment of Alzheimer's disease
(ASGCT 2026)
- "Conclusion Taken together, the data collected to date demonstrate that AVB-406 is a well- tolerated tau-targeting miRNA with a novel cross-species BBB-penetrant capsid, achieving robust, dose-dependent and sustained MAPT knockdown with CNS specificity in preclinical models. These findings support AVB-406 as a promising one-time, IV treatment approach for AD and other tauopathies with potential for clinical translation"

Development of a scalable platform production process for AVB-406, a blood–brain barrier–crossing AAV vector for MAPT knockdown in Alzheimer’s disease
(ASGCT 2026)
- "The final material exhibited high purity with high-quality packaged DNA. Importantly, the purified AVB-406 retained its expected potency, demonstrating the ability to cross the blood-brain barrier and to knock down endogenous MAPT mRNA."

Development and translational validation of AVB-406: an intravenous AAV-delivered miRNA targeting MAPT for the treatment of Alzheimer's disease
(ASGCT 2026)
- "AVB-406 was well tolerated in both murine models and non-human primate studies, supporting clinical translation. These data establish AVB-406 as a promising therapeutic candidate for Alzheimer's disease and demonstrate the capabilities of the vMiX™ platform for systematic miRNA therapeutic development, enabling rapid progression from target identification through preclinical validation and clinical-scale manufacturing."

AviadoBio Presents Positive Preclinical Data for AVB-406 Program in Alzheimer’s Disease and Other Tauopathies at ASGCT 2026
(Yahoo Finance)
- "The data demonstrate progression of AVB-406 across preclinical validation, platform discovery and scalable manufacturing, and collectively support the company’s plans to advance AVB-406 into clinical trials for AD by the end of 2026...In a series of preclinical studies, including human iPSC models and mice, AVB-406 was evaluated for target engagement, biodistribution, dose-responsive activity, tolerability, and efficacy; Notably, AVB-406 sustained dose-dependent MAPT mRNA knockdown in brain cortex tissue in humanized mice, including in key brain regions associated with AD pathology, with in situ hybridization confirming neuronal specificity of miRNA guide expression....AVB-406 manufacturing at 3L and 10L scales yielded consistent recovery aligned with industry standards, further supported by a 15L toxicology batch achieving >30% recovery."
