Crosstalk Between Dopamine and the Epigenetic Enzyme LSD1/KDM1a: Implications for Amphetamine Response.
(PubMed, J Neurochem)
- "On the other hand, while a single dose of the AMPH did not alter uLSD1 and neuroLSD1 isoforms' expression, repeated AMPH administration led to a transient increase followed by a reduction of neuroLSD1 transcripts' abundance in the striatum and hippocampus. In conclusion, our data reveal a critical interplay between dopaminergic neurotransmission and the expression of LSD1 isoforms in the brain, highlighting their potential role in modulating dopamine-dependent behaviors."

Adolescent stress differentially modulates the affective, psychomotor, and neural responses to a first amphetamine exposure in male Wistar rats.
(PubMed, Pharmacol Biochem Behav)
- "Conversely, the expression of Rho GTPase-activating protein 32 (P250gap), cAMP-response element binding protein (Creb), and DNA methyltransferase 3A (Dnmt3a) was cross-sensitized in the NAc. The coexistence of cross-sensitization and cross-tolerance neurobehavioral effects between CUS and amphetamine supports the idea that ELA can simultaneously blunt and heighten different brain substrates, collectively increasing the risk of drug dependence."

Real-Time In Vivo Monitoring of Cholinergic Neurotransmission in the Mouse Brain Using a Microelectrochemical Choline Biosensor.
(PubMed, Eur J Neurosci)
- "Systemic administration of donepezil produced a pronounced decrease in current in both the prefrontal cortex and hippocampus, with scopolamine and amphetamine resulting in signal increases that were not observed in animals with selective saporin lesioning (murine-p75) of the cholinergic basal forebrain. Furthermore, continuous biosensor recording in both regions displayed diurnal oscillations across repetitive light-dark phases. All are consistent with successful monitoring of endogenous changes in cholinergic neurotransmission."

Individual Differences in Frequency Modulated 50-kHz Ultrasonic Vocalizations Reflect but Do Not Predict Cocaine Self-Administration in Rats.
(PubMed, Behav Brain Res)
- "Twenty singly housed rats underwent behavioral testing (elevated plus maze, novel object recognition, and tickling-induced vocalizations) and amphetamine sensitization...However, anticipatory FM 50-kHz USVs do not provide a reliable indicator of SA intensity. Accurate interpretation of FM 50-kHz USVs requires integration with behavioral and neurobiological markers to effectively identify addiction risk phenotypes."

A Rare Presentation of Gastric Ischemia Manifesting as Gastric Pneumatosis in the Context of Amphetamine Use
(ACG 2025)
- "A nasogastric tube was placed for decompression, and the patient was started on ceftriaxone and metronidazole. Our case highlights the need to consider gastric ischemia in patients with amphetamine use disorder presenting with nonspecific gastrointestinal symptoms and imaging suggestive of gastric pneumatosis. Early recognition of gastric pneumatosis is crucial to improve patient outcomes, as it may indicate ischemia and carries a risk of increased mortality.Figure: Axial computed tomography image of the abdomen demonstrating gastric pneumatosis, indicated by red arrows."

Beyond Varices: Left Gastric Pseudoaneurysm as a Rare Cause of Upper GI Bleed in Cirrhosis
(ACG 2025)
- "Several interventions, including hemostasis powders, epinephrine, and ethanolamine, were unsuccessful, so a fully-covered stent was placed to tamponade the bleed. It is possible that the pseudoaneurysm developed due to severe local inflammation from the amphetamine-induced esophagitis. Worsening esophagitis and coagulopathy likely caused the pseudoaneurysm to rupture, resulting in a bleed refractory to endoscopic intervention and requiring embolization.Figure: Figure 1: CT Angiography Demonstrating the Pseudoaneurysm in the Gastroesophageal Branch of the Left Gastric Artery"

Revisiting the Role of Serotonin in ADHD
(AACAP 2025)
- "In addition, several medications that positively impact ADHD symptoms (eg, amphetamine, viloxazine, and monoamine oxidase [MAO] inhibitors) are associated with enhanced serotonergic activity...Of note, human fMRI studies indicate that fluoxetine challenge produces changes in activation within the prefrontal cortex similar to what is seen with ADHD medications (Chantiluike et al, 2015). We conclude that 5-HT has an important role in key associated features of ADHD and indirectly contributes to the regulation of core features of the disorder. Implications for treatment will be discussed."

To Stimulate or Supplement: A Nationwide Review of Long-Term Outcomes in Patients With ADHD
(AACAP 2025)
- "Current AACAP guidelines include stimulants such as methylphenidate or amphetamine-based medications as first-line therapy. In this cohort study, combined therapy was associated with increased risks of MDD, GAD, CD, and SI compared to stimulants or supplements alone. Interestingly, compared to supplement monotherapy, stimulant monotherapy was associated with increased risks of MDD, GAD, and CD. Further study is needed to clarify the effects of supplements in mental health outcomes of patients with ADHD, including interactions with stimulant medications.EBP, STIM, MAE"

Inducing Striatal Overgrowth in Mice to Model Autism-Linked Neurodevelopmental Changes
(AACAP 2025)
- "Adult offspring completed a behavioral battery including open field, rotarod motor learning, water T-maze habit and reversal learning, and amphetamine (AMPH)-induced stereotypy assays... These findings provide the first direct evidence that prenatal activation of mGluR5 via CHPG ICV injection induces striatal cellular overgrowth and leads to enduring changes in striatal-dependent behavior. This model offers a tractable platform for exploring mechanisms linking early striatal overgrowth to ASD-relevant behavioral phenotypes and may inform future therapeutic targets.ASD, NEURODEV, ANI"

Optimizing ADHD Medication Management: From Stimulants to Nonstimulants in Children and Adolescents
(AACAP 2025)
- "Specifically, the presentation will cover dosing regimens, side effects, and monitoring considerations for CNS stimulants (eg, methylphenidate, amphetamine salts), alpha-2 adrenergic agonists (eg, guanfacine, clonidine), and NRIs (eg, atomoxetine, viloxazine). CNS stimulants remain the first-line treatment for ADHD, with a 70%-80% response rate in children... Effective ADHD management requires a personalized approach, balancing medication efficacy with tolerability. Clinicians must closely monitor side effects and ensure that treatment aligns with the patient's developmental stage, comorbidities, and individual needs. Continued collaboration with families and frequent follow-up are critical to optimizing long-term outcomes for children and adolescents with ADHD.ADHD, PPC, TREAT"

Molecular basis of vesicular monoamine transport and neurological drug interactions.
(PubMed, Cell Rep)
- "Here, we report cryo-electron microscopy structures of human VMAT2 capturing a cytoplasmic-open state with reserpine and lumenal-facing states with serotonin and histamine in open, amphetamine in less open, tetrabenazine in fully occluded, and unbound VMAT2 in partially occluded conformations. Liposome-based analyses demonstrate that amphetamine directly induces the release of a fluorescent monoamine analog via VMAT2, consistent with an exchange mechanism underlying psychostimulation. These findings reveal the molecular basis of monoamine storage and drug interactions by VMAT2, informing therapeutic development for neurological diseases and substance abuse."

The Effect of Intraocular Haloperidol on Motor Function in Models of Two Neuropsychiatric Disorders: Implications for the Origin and Treatment of Parkinson's Disease, Psychosis and Drug Addiction.
(PubMed, Brain Sci)
- " Motor tests included open field performance and the latency to perform three motor tests after the IVIT injection of haloperidol with and without amphetamine pretreatment. These results illustrate that IVIT injections of DA and melatonin receptor antagonists can differentially alter motor function via the retina. This suggests that the retina may be a treatment target not only for PD but also for other DA- and melatonin-mediated disorders such as drug addiction, psychosis and schizophrenia."
