Stimulant Medications and Sleepiness in ADHD: Insights from a Sleep Deprivation Study
(WSS 2025)
- "Stimulant medications, such as Methylphenidate and Amphetamine, effectively mitigate excessive sleepiness experienced by young men diagnosed with ADHD, even under conditions of extended sleep deprivation. These findings have important clinical and theoretical implications. Given the negative effects of sleepiness on cognition and emotional regulation, these results suggest that the beneficial effects of stimulant medications in ADHD may partially stem from their effect on sleepiness."

Treatment Considerations in Co-morbid Schizophrenia and Narcolepsy: A Case Report
(WSS 2025)
- "Some alertness promoting medications used in narcolepsy treatment, such as modafinil, methylphenidate and amphetamine, may exacerbate psychosis by increasing dopamine activity...He was previously intolerant to modafinil (emotional numbing) and Concerta (appetite suppression) as per chart review.Paliperidone was effective for psychosis but worsened sleepiness, which was disabling... In conclusion, some traditional stimulants may worsen psychosis in PWN. Pitolisant, an inverse H3 agonist, which does not theoretically affect dopamine pathways, may be a suitable treatment for sleepiness and cataplexy in comorbid psychosis."

Evidence of Nose-To-Brain Delivery of Lithium: Implications for Mood Disorders and Beyond
(WFSBP 2025)
- "(3) in vivo rat model showed that intranasal lithium delivery produced same behavioural response than intraperitoneal lithium delivery in an amphetamine model of mania... This study provides the first evidence of successful nose-to-brain delivery of lithium using nanoparticle network hydrogels. The results also suggest that intranasal lithium can exert significant behavioural changes with significantly less circulating serum levels. These results have important implications for translational research in mood disorders, given the need for long-term treatments with potential for systemic side effects."

Intraday trends of chemical biomarkers in wastewater monitored through automated real-time surveillance.
(PubMed, Water Res)
- "Population biomarkers exhibited distinct temporal profiles: paraxanthine remained stable during working hours, while candesartan declined even at peak population times - highlighting implications for normalization, particularly with day visitors...During the special events, MDMA, ketamine, and amphetamine loads rose with distinct profiles - one event showing a gradual overnight rise, the other a morning peak...Levamisole loads rose on weekends and during events, mirroring benzoylecgonine, indicating continued presence as harmful cocaine adulterant. Automated analysis reveals short-term intraday trends, providing additional insights into the sources of chemical marker loads - such as consumption, excretion, and contributions from transient populations - that are typically averaged out in standard 24-hour composite samples."

A Rare Presentation of Gastric Ischemia Manifesting as Gastric Pneumatosis in the Context of Amphetamine Use
(ACG 2025)
- "A nasogastric tube was placed for decompression, and the patient was started on ceftriaxone and metronidazole. Our case highlights the need to consider gastric ischemia in patients with amphetamine use disorder presenting with nonspecific gastrointestinal symptoms and imaging suggestive of gastric pneumatosis. Early recognition of gastric pneumatosis is crucial to improve patient outcomes, as it may indicate ischemia and carries a risk of increased mortality.Figure: Axial computed tomography image of the abdomen demonstrating gastric pneumatosis, indicated by red arrows."

Beyond Varices: Left Gastric Pseudoaneurysm as a Rare Cause of Upper GI Bleed in Cirrhosis
(ACG 2025)
- "Several interventions, including hemostasis powders, epinephrine, and ethanolamine, were unsuccessful, so a fully-covered stent was placed to tamponade the bleed. It is possible that the pseudoaneurysm developed due to severe local inflammation from the amphetamine-induced esophagitis. Worsening esophagitis and coagulopathy likely caused the pseudoaneurysm to rupture, resulting in a bleed refractory to endoscopic intervention and requiring embolization.Figure: Figure 1: CT Angiography Demonstrating the Pseudoaneurysm in the Gastroesophageal Branch of the Left Gastric Artery"

Assessment of risk factors associated with methadone-induced deaths: A population study based on forensic autopsies (2003-2022).
(PubMed, Forensic Sci Int)
- "This forensic study reveals that poly-drug use is a major contributor to methadone-related fatalities. Concomitant use of methadone with benzodiazepines or psychostimulants is particularly hazardous. Pharmacological interactions must be carefully assessed in methadone users. Our findings underscore the value of forensic data in identifying lethal drug combinations and supporting safer prescribing practices and public health strategies."

Gold-Containing Compound Auranofin Ameliorates 6-OHDA Induced Neurotoxicity in Rats: Via Targeting PI3K/Akt/GSK-3β Signalling.
(PubMed, Clin Exp Pharmacol Physiol)
- "After performing the amphetamine challenge test, rodents were treated with AUF (10 mg/kg) and AUFHNPs (5 and 10 mg/kg) for 21 days...It also restored morphological changes and increased expression of pPI3K/pAKT, followed by inhibition of GSK-3β. Thus, the AUFHNPs were more significant than AUF alone and exerted neuroprotection via modulation of PI3K/AKT/GSK-3β signalling pathways."

Trace amine-associated receptor 1 agonists differentially regulate dopamine transporter function.
(PubMed, Mol Pharmacol)
- "RO5166017, but not RO5256390 or ulotaront, increased amphetamine-induced dopamine efflux in a TAAR1-dependent manner. All 3 affected DAT transport and/or trafficking, with each exhibiting a unique profile of direct and heterologous effects, some of which were TAAR1-dependent. These issues should be considered with therapeutic design and clinical use of TAAR1 agonists."

Using a rich-lean transition procedure to evaluate putative antianxiety medications.
(PubMed, J Pharmacol Exp Ther)
- "Acute administration of benzodiazepines (midazolam, alprazolam) and an imidazotriazine (TPA-023B) selectively reduced pausing in rich-lean transitions, a behavioral effect consistent with clinically effective antianxiety drugs in other preclinical anxiolysis procedures. Morphine and (+)amphetamine selectively increased rich-lean pausing, an effect consistent with an anxiogenic response in other anxiolysis procedures...SIGNIFICANCE STATEMENT: Transitions from favorable to unfavorable (ie, rich-lean) schedules of reinforcement reliably disrupts operant behavior, and these disruptions can be ameliorated via benzodiazepine administration. Our results indicate that the rich-lean transition procedure has utility in the assessment of putative antianxiety medications."
