Single-Dose siRNA Therapeutic Targeting Angiotensinogen Produces Durable Blood Pressure Reduction in Spontaneously Hypertensive Rhesus Monkeys (ASGCT 2026) - "Twenty spontaneously hypertensive rhesus monkeys were randomized based on baseline blood pressure to receive a single subcutaneous dose of MWX401 (1.5, 5, or 15 mg/kg), placebo, or daily oral valsartan (2.67 mg/kg) as a positive control. Conclusion These data demonstrate that a single subcutaneous dose of MWX401, an RNA-based therapeutic targeting AGT, can achieve long-lasting blood pressure control in a clinically relevant non-human primate model. The magnitude, durability, and dose dependence of both AGT suppression and blood pressure reduction support MWX401 as a promising long- acting RNA therapeutic for hypertension and provide a strong rationale for infrequent dosing strategies in clinical development."
