Guanfacine for Dexmedetomidine Withdrawal (SCCM 2026) - "Historically, clonidine has been used for this purpose, however its use is limited by cardiovascular effects, namely hypotension... The results of this evaluation reveal a lack of standardized guanfacine dosing strategies in dexmedetomidine weaning, suggesting an opportunity for protocol development. Additionally, the findings suggest an education opportunity for providers on ensuring the appropriate guanfacine formulation is ordered for the planned route of administration. Immediate future directions for this project include a research project designed to evaluate the comparative efficacy of dexmedetomidine weaning strategies."

Extended-Release Viloxazine for Children and Adolescents With Attention Deficit Hyperactivity Disorder. (PubMed, J Pediatr Pharmacol Ther) - "Non-stimulant medication therapy for children, adolescents, and young adults with attention-deficit/hyperactivity disorder (ADHD) has included alpha 2-agonists (guanfacine and clonidine) as well as a norepinephrine reuptake inhibitor (atomoxetine) for multiple years...Extended-release viloxazine (SPN-812) has been shown in phase 2 and phase 3 trials to be an effective, well-tolerated alternative for some children with ADHD...Although continued evaluation to confirm the reduced symptoms of ADHD in children along with the safety profile is needed, extended-release viloxazine may offer a once-a-day pharmaceutical treatment option for patients in which stimulant medication is not effective or not a favorable option. It may also be beneficial for those children and adolescents with a comorbidity of depression."

Safety and efficacy of guanfacine extended-release in adults with attention-deficit/hyperactivity disorder: an open-label, long-term, phase 3 extension study. (PubMed, BMC Psychiatry) - "There were no major safety concerns during long-term GXR administration in adults with ADHD. After long-term treatment, patients had significant improvements from baseline in ADHD symptoms, QoL, and executive functioning."

Guanfacine-Extended Release for Treatment of Adults with Adhd (APSARD 2020) - "GXR may be useful for adults to address ADHD-related impairments such as difficulty in falling asleep, excessive irritability or impulsivity with or without concurrent stimulants. More research is needed to explore the use of GXR for adults with ADHD."
