Dimerization of the S. cerevisiae Spo11 core complex. (PubMed, bioRxiv) - "Here, we investigated the dimerization of S. cerevisiae Spo11 in complex with its partners Ski8, Rec102, and Rec104...Finally, we found that Rec102 exerts a key DNA-binding function, essential for catalysis, and show that it also participates in dimerization through trans contacts with Ski8. Our work provides new insights into the mechanism of Spo11 dimerization and the role of its partners in initiating meiotic recombination."

Cryo-EM structures of the Spo11 core complex bound to DNA. (PubMed, Nat Struct Mol Biol) - "We report cryo-electron microscopy structures at up to 3.3-Å resolution of DNA-bound core complexes of Saccharomyces cerevisiae Spo11 with Rec102, Rec104 and Ski8. In these structures, monomeric core complexes make extensive contacts with the DNA backbone and with the recessed 3'-OH and first 5' overhanging nucleotide, establishing the molecular determinants of DNA end-binding specificity and providing insight into DNA cleavage preferences in vivo. The structures of individual subunits and their interfaces, supported by functional data in yeast, provide insight into the role of metal ions in DNA binding and uncover unexpected structural variation in homologs of the Top6BL component of the core complex."

Assessing kidney function in pregnancy: gestation specific centile reference ranges for serum creatinine, urea, cystatin c and beta-2-microglobulin (UKKW 2024) - " Prospective cohort study of women receiving antenatal care at two UK tertiary hospitals (2018-2021) recruited to Pregnancy Adaptations in Renal disease study (REC15/WA/0009) and Prediction of pregnancy complications study (REC02-03-033)... 539 participants were included. 444(82.4%) had an uncomplicated pregnancy and delivery and 95(17.6%) developed a risk factor for AKI (Table 1). Table 2 describes means for all biomarkers at each gestational window by sub-group."

In this ethnically diverse population, mean and median serum creatinine values were lower than previous reports in population cohorts. Serum creatinine above 63umol/l at any gestation of pregnancy is above the 97th centile and may represent pathology. Urea values above 5.5umol/l in the first and third trimester, and 4.6mmol/l in the second warrant further assessment."

(PubMed, Nat Struct Mol Biol) - "We provide molecular analysis of Saccharomyces cerevisiae Spo11 purified with partners Rec102, Rec104 and Ski8...Mutations that reduce DNA binding in vitro attenuate DSB formation, alter DSB processing and reshape the DSB landscape in vivo. Our data reveal structural and functional similarities between the Spo11 core complex and Topo VI, but also highlight differences reflecting their distinct biological roles."
