Neuropharmacological potential of Mimosa tenuiflora in adult zebrafish: An integrated approach to GABAergic and serotonergic neuromodulation.

- "The anxiolytic response induced by alcohol withdrawal and the seizure induced by pentylenetetrazole were also tested. The possible mechanisms of anxiolytic and antidepressant actions of FATEM were evaluated through the administration of specific antagonists (Flumazenil, Cyproheptadine, Pizotifen or Granisetron)...FATEM did not present acute toxicity and caused a reduction in locomotor activity (p < 0.0001 vs. Control) similar (p< 0.0001) to Diazepam, indicating a sedative/anxiolytic effect...FATEM also prevented anxious behaviors induced by alcohol withdrawal and exhibited an antidepressant effect in the ZTI (p < 0.0001 vs. Control) similar (p < 0.0001) to the effect of Fluoxetine, which was reversed by serotonergic antagonists. In silico evaluations indicated favorable pharmacokinetic properties and affinity of FATEM with GABAergic and serotonergic receptors. - "Propranolol, metoprolol, candesartan, topiramate, divalproex, lisinopril, amitriptyline, and venlafaxine have the strongest evidence to support for use. Flunarizine and pizotifen may also be effective. Selection of preventive treatments is based on individual characteristics, comorbid conditions, efficacy, contraindications, side effects, cost, compliance, and drug. An adequate trial of migraine prophylaxis is usually 2 months at the target dose, and it is always important to re-evaluate indication for prophylactic use after a period of time."

Furthermore, high dose of pizotifen (5μg) weakly affected motor performance and did not influence the locomotor activity in normal animals. In summary, our findings suggest that pizotifen strengthens the inhibitory synaptic transmission and exerts antinociceptive effect on both neuropathic pain and inflammatory pain in the spinal cord, and may serve as a promising remedy for chronic pain."

- "Recent large trials of established conventional pharmaceuticals such as propranolol, pizotifen, topiramate and amitriptyline for childhood migraine have failed, but the use of a quasi-placebo in future trials could help. We reviewed the evidence for angiotensin antagonists including candesartan in adults, but found a lack of evidence for their use in children. There have been new developments in pharmaceuticals recently, including a more selective 5-HT1F agonist, lasmiditan, an effective acute treatment with no vasoconstrictor activity in adults, currently being tested in children...So, there has been much progress, but mostly for adults. We are in urgent need of more migraine trials in children."

Anxiolytic-like effect of natural product 2-hydroxy-3,4,6-trimethoxyacetophenone isolated from Croton anisodontus in adult zebrafish via serotonergic neuromodulation involvement of the 5-HT system.

- "Pentylenetetrazole (PTZ) was used to induce seizure by immersion...The higher doses (3 and 10 mg/kg; v.o.) produced signs of anxiolytic action in the light/dark test, and this effect was reversed by the pizotifen (antagonist 5HTR and 5HTR), having the potential to form a complex with 5HTR1B. However, the anxiolytic effect of HTMCX has not been abolished by flumazenil (antagonist GABA), cyproheptadine (antagonist 5HTR), and granisetron (antagonist 5HTR). Therefore, HTMCX demonstrated an anxiolytic effect, suggesting that the 5HTR and 5HTR receptors may be involved in the pharmacological performance of this acetophenone in the central nervous system."
