Factors influencing time to viral rebound during analytical treatment interruptions in HIV cure trials (AIDS 2024) - "The trials investigated the following interventions alone or in combination: broadly neutralizing antibodies (bNAb) (3BNC117, 10-1074), histone deacetylase inhibitors (HDACi) (romidepsin, panobinostat), HIV-1 peptide vaccine (Vacc-4x) and toll-like receptor 9 (TLR9) agonist (MGN1703). Our findings shed new light on factors influencing time to viral rebound as well as loss of virologic control. These findings can inform the design of novel cure trials but also highlight the potential impact of early bNAb treatment on virological control during ATI."

NK CELL BIOMARKERS THAT PREDICT TIME-TO-REBOUND IN HIV+ INDIVIDUALS UNDERGOING ATI (CROI 2023) - " We analyzed samples from two ATI cohorts: the REDUC cohort, which consisted of 17 participants that received therapeutic vaccination (Vacc4x) with the HDAC inhibitor Romidepsin prior to ATI, and the ACTG A5345 cohort, which consisted of 33 chronic- and 12 acute-treated participants that underwent ATI in the absence of any intervention. As CD56-CD16+ NK cells have been associated with bnAb production, these results suggest a potential role of NK cells and HIV-specific antibodies in control of viral replication. Siglec-7+ NK cells may temporarily control viral rebound during ATI. The findings with CXCR6 and CD49a implicate a role for memory NK cells in the preserved immune responses found in acute-treated individuals."

Characterization of the HIV-1 transcription profile after romidepsin administration in ART-suppressed individuals. (PubMed, AIDS) - "In these patients, romidepsin increased early events in HIV transcription (initiation and especially elongation), but had less effect on later stages (completion, multiple splicing) that may be required for comprehensive latency reversal and cell killing. Without cell death, increased HIV transcription before or after latency reversal may hasten viral rebound after therapy interruption."
