Subcutaneous administration of isatuximab by an on-body delivery system (OBDS) in multiple myeloma patients: results of a nurse survey

- P1 | "Introduction: Intravenous (IV) isatuximab (Isa) in combination with pomalidomide-dexamethasone (Pd) is approved for the treatment of relapsed/refractory multiple myeloma (RRMM) patients...Extent of drug delivered not clear in case of interruption (83%) and need to press/hold a button to pause injection (67%) were agreed upon as potential disadvantages for HCPs; risk of allergy to the adhesive tape (50%) was reported as a potential disadvantage for patients, although no interruptions or allergy were observed in 581 administrations via OBDS in the Phase 1b study (IMS 2023; P-306). Findings from this nurse survey show a high level of confidence and preference for SC Isa treatment delivery using a hands-free OBDS vs IV administration, due to its ease of use, tolerability, and time savings, suggesting its applicability in routine clinical practice."

- "Among the species studies, only rat CaMKIIα with pT286/pT305/pT306 exhibited kinase domain oligomerization. Furthermore, we revealed that the sensitivity of CaMKII to PP2A in the three species differs, with rat, C. elegans, and hydra being less dephosphorylated in that order. The evolutionarily acquired features of mammalian CaMKIIα-specific structural arrangement and phosphatase tolerance may differentiate neuronal function between mammals and other species."

- "Using genome sequence data from C. elegans MA lines carrying a mutation (mev-1) that increases the cellular titer of reactive oxygen species (ROS), leading to increased oxidative stress, we find the base-substitution spectrum is similar between mev-1, its wild-type progenitor (N2), and another set of MA lines derived from a different wild strain (PB306)...Further, the mutational properties of mononucleotide repeats in all strains are different from those of nonmononucleotide sequence, both for indels and base-substitutions, and whereas the nonmononucleotide spectra are fairly similar between MA lines and wild isolates, the mononucleotide spectra are very different, with a greater frequency of A:T → T:A transversions and an increased proportion of ±1-bp indels. For amines, 1,3-substitution (vs 1,4) and aromaticity were both found to be beneficial to chemical performance."

- "The Chk1 inhibitor SAR-020106 is a potent sensitizer for DNA damage-induced cell death in glioblastoma therapy strongly reducing clonogenicity of tumour cells. Selectively enhanced p53-mut cell death may provide stronger responses in tumours defective of non-homologous end joining (NHEJ). Our results suggest that a multimodal therapy involving DNA damage inducers and DNA repair inhibitors might be an effective anti-tumour strategy with a low risk of neurotoxicity."

Collectively, these findings indicate that, conversely to human FVa, APC-proteolysis of ptFVa at Arg306 or deletion of the unique disulfide bond does not abrogate cofactor function. Concomitantly, the structural integrity of ptFVa is stabilized, preventing A2-domain dissociation, thereby providing productive interactions. Additional support comes from plasmin-mediated proteolysis, suggesting an enhanced stability of essential functional regions in ptFVa ."
