Hydrochlorothiazide and chlorthalidone use and glaucoma risk: pharmacovigilance analysis and nationwide cohort study.
(PubMed, Front Pharmacol)
- "Although the FAERS pharmacovigilance suggested a potential glaucoma risk with hydrochlorothiazide and chlorthalidone, population-based validation found no significant association. Pharmacovigilance signals should be confirmed with population-based data to identify true drug-glaucoma associations."

A RARE CASE OF MYOGLOBIN NEPHROPATHY CAUSED BY PARANEOPLASTIC DERMATOMYOSITIS SECONDARY TO SMOLDERING MULTIPLE MYELOMA
(ISN-WCN 2026)
- "These findings were suggestive of dermatomyositis. The only comorbidity was hypertension for which he was taking a combination tablet consisting of 10 mg amlodipine, 160 mg valsartan and 12.5 mg hydrochlorothiazide...9 This case serves as a clear example of its utility in early diagnosis. This patient was referred to a haem-oncologist and received 6 cycles of bortezomib/leflunamide and dexamethasone...This prompted further investigation into the type of protein in the urine. This case was referred to a nephrologist and highlights how urine analysis can be the harbinger of diagnosis of a systemic illness and must be done routinely in the form of a simple bedside urine dipstick test and not underestimated"

DISTAL NEPHRON RESPONSE FOLLOWING THIAZIDE TREATMENT IN A RODENT MODEL OF PRESSURE OVERLOAD-INDUCED HEART FAILURE
(ISN-WCN 2026)
- "TAC mice were then assigned to receive either vehicle or hydrochlorothiazide (HCTZ) in their drinking water for 5 weeks. Results 7 weeks after surgery, TAC induced cardiac hypertrophy in high-salt fed mice, which was attenuated by HCTZ treatment...Consistent with RAAS activation, gene expression of pendrin was also significantly increased by HCTZ treatment. Furthermore, changes in the expression of genes associated with the activation of α-ketoglutarate-OXGR1 pathway, which is implicated in alkalosis-dependent pendrin expression, were observed in the HCTZ-treated group. Conclusion In heart failure, thiazide therapy may induce counter-activation of distal nephron sodium chloride reabsorption mechanisms via RAAS activation and pathways associated with alkalosis, which potentially lead to treatment resistance."

THE EFFECTS OF THIAZIDES ON EGFR CHANGES IN PATIENTS WITH ADVANCED CHRONIC KIDNEY DISEASE
(ISN-WCN 2026)
- "This study aimed to investigate the effects of thiazides on renal function decline in patients with advanced CKD.Methods This was a single-center retrospective study of patients diagnosed with CKD G4 (eGFR 15–29 mL/min/1.73 m2) or G5 (eGFR <15 mL/min/1.73 m2) who initiated thiazides (trichloromethiazide, hydrochlorothiazide, or indapamide) from January 2014 to September 2024 in the Department of Nephrology, Kansai Electric Power Hospital in Japan. In this study, thiazides in patients with advanced CKD was associated with a slower decline in eGFR, suggesting their potential usefulness in managing advanced CKD. Furthermore, a reduction in proteinuria was observed in addition to antihypertensive effects, supporting the hypothesis that thiazides may have reno-protective effects by attenuating glomerular overload."

A HIDDEN HAZARD OF HEPARIN: LIFE-THREATENING HYPONATREMIA AND HYPERKALEMIA DUE TO TYPE IV RTA
(ISN-WCN 2026)
- "He subsequently received unfractionated heparin for thromboprophylaxis and dexamethasone for cord compression...Hypertonic saline, sodium zirconium cyclosilicate, and fludrocortisone were administered, and heparin, lisinopril, and hydrochlorothiazide were discontinued, resulting in rapid normalization of serum sodium, potassium, bicarbonate, and blood pressure. Results Download: Download high-res image (159KB)Download: Download full-size imageConclusion Although heparin associated Type 4 RTA is a well-known pharmacologic effect, its clinical impact is often underestimated...This case highlights that even routine heparin prophylaxis can cause severe aldosterone deficiency. Prompt recognition and discontinuation of heparin, along with temporary mineralocorticoid replacement, can be lifesaving."

Uncovering redox-specific biotransformation of organic micropollutants.
(PubMed, J Hazard Mater)
- "While some OMPs were well-removed (e.g., paracetamol and hydrochlorothiazide) or persistent (e.g., diglyme and bentazon) under all tested conditions, the fate of most OMPs was redox-dependent in both rate and progress along the degradation pathway...Our results provide a more comprehensive view of OMP fate and TP formation across redox conditions. These insights can support the optimization of OMP removal strategies in engineered and natural redox-shifting systems."

Temporal Risk of Non-Melanoma Skin Cancer with Lisinopril and Hydrochlorothiazide as Monotherapy and in Combination: A Retrospective Cohort Study Using the TriNetX Database
(AAD 2026)
- "Z-scores between cohorts identified HCTZ alone to carry a significantly greater risk of cSCC at 5 years [RR=3.01 (2.15, 4.213), p<0.0001] when compared to combination LIS+HCTZ. Our study demonstrates that LIS and HCTZ either taken in combination or as monotherapy for HTN increases the risk of NMSC development."

A Comparison of the Protective effects of Systemic vs Oral Retinoids on Non Melanoma Skin Cancers in Acne Patients: A Retrospective Cohort Study
(AAD 2026)
- "Two matched cohorts were created: systemic retinoids (alitretinoin, isotretinoin, etretinate, acitretin; n=34,851) and topical retinoids (adapalene, tazarotene, bexarotene, tretinoin; n=34,851). Patients with known NMSC risk factors (chemical exposures, HPV, or use of BRAF inhibitors, cyclosporine, voriconazole, hydrochlorothiazide) were excluded... The enhanced protective effect of systemic retinoids may reflect their deeper influence on keratinocyte turnover and skin cell regeneration compared to topical formulations. Further studies are warranted to clarify the relative efficacy of individual agents and optimize retinoid prophylaxis in high-risk patients."

Do Not Dismiss Incidental Hyperkalemia in Childhood: Early Recognition of Pseudohypoaldosteronism Type II.
(PubMed, Cureus)
- "Treatment with hydrochlorothiazide (HCTZ) resulted in normalization of serum potassium and correction of the metabolic acidosis. Genetic testing revealed a heterozygous KLHL3 variant (c.1501C>T, p.Pro501Ser), confirming the diagnosis of PHAII. This case highlights the importance of not dismissing incidentally detected hyperkalemia in children and illustrates the value of performing a structured evaluation that includes confirming the persistence of hyperkalemia, acid-base assessment, urinary potassium analysis, and family history-taking to facilitate early diagnosis of renal tubular disorders such as PHAII."

EARLY-ONSET, ASYMPTOMATIC NIFEDIPINE HEPATOTOXICITY: EXPANDING THE SPECTRUM OF CALCIUM CHANNEL BLOCKER LIVER INJURY
(ACC 2026)
- "He was started on intravenous nicardipine, later transitioned to oral nifedipine 90 mg daily plus losartan...Nifedipine was discontinued, replaced with hydrochlorothiazide, and liver enzymes normalized within 90 days... This case demonstrates that nifedipine can cause early, asymptomatic hepatocellular injury even in young, otherwise healthy individuals. Clinicians should maintain vigilance for unexplained transaminase elevations soon after initiation, and promptly discontinue the drug to prevent progression."

A PRICE TO COIL: SECONDARY HYPERTENSION AFTER BILATERAL ACCESSORY RENAL ARTERY EMBOLIZATION
(ACC 2026)
- "He was treated with intravenous clevedipine followed by amlodipine and hydrochlorothiazide... Bilateral accessory renal artery coiling during endovascular aneurysm repair can cause loss of renal perfusion and downstream activation of the RAAS leading to abrupt secondary hypertension, even when the main renal arteries and graft remain patent. Careful preoperative assessment and close postoperative monitoring are essential to recognize and manage this complication."

ARE EDEMA EVENTS IN PRECISION ASSOCIATED WITH AN INCREASED RISK OF HOSPITALIZATION FOR HEART FAILURE?
(ACC 2026)
- "As a result, some had to switch from loop diuretics or ≥1 diuretic to hydrochlorothiazide (HCTZ; 25 mg/d). HFH in pts receiving aprocitentan was rare and mainly occurred in pts with pre-existing HF and other comorbidities. The protocol-mandated switch to HCTZ from other diuretics most probably contributed to HFH. The incidence of edema events was similar in both HF and non-HF groups, but HFH was higher in the HF cohort, suggesting HFH was not primarily driven by edema observed in PRECISION."

Pharmacogenetics of RAS-affecting AGT and ACE variants and the efficacy of Valsartan/HCTZ therapy.
(PubMed, Sci Rep)
- "Considerable inter-individual variability has been observed in the blood pressure response to valsartan/hydrochlorothiazide (Valsartan/HCTZ), and genetic differences within the renin-angiotensin system may contribute to this heterogeneity...A significant AGT rs5050 × ACE I/D interaction revealed the greatest reduction in AC/II (20.6 ± 2.3 mmHg) and the lowest in AA/DD (3.80 ± 2.33 mmHg). These findings highlight meaningful multilocus effects and support the potential for genotype-guided antihypertensive therapy."

Beads of Burden: Severe Hypercalcemia Following Orthopedic Calcium Sulfate Beads Implantation
(NKF-SCM 2026)
- "METHODS/CASE SUMMARY A 72-year-old man with hypertension on hydrochlorothiazide and GERD on calcium carbonate (Tums) underwent revision right total knee arthroplasty (Day 0)...He received Plasmalyte A (calcium-free) and a single dose of zoledronic acid on Day 5, after which calcium gradually normalized (≈9.8 mg/dL by Day 11) with concurrent improvement in renal function...CONCLUSION This case underscores a rare but clinically important complication of Stimulan calcium-sulfate beads. Perioperative monitoring of serum calcium and renal function, preoperative medication review, judicious bead use, and early recognition allows prompt intervention and reversal."
