Ocular Safety and Efficacy of AAV-mediated Tyrosinase Gene Augmentation in a Nonhuman Primate Model.
(PubMed, bioRxiv)
- "A novel AAV2-based capsid (ATX002) was packaged with the human VMD2 promoter and TYR (h TYR ) fused with mGreenLantern (mGL)...Subretinal AAV- TYR delivery is a safe and effective approach with the potential to induce RPE pigmentation. These findings support the use of AAV- TYR gene therapy for OCA1, demonstrating efficacy and a manageable safety profile in a large-animal model, and provide a critical bridge toward human clinical translation."

Manufacturability of ATX002, a Next Generation Ophthalmologic AAV Capsid
(ASGCT 2026)
- "Minimal to no process optimization work would be required for any of the platforms evaluated to be commercial manufacturing ready, further derisking (or ultimately not requiring) any late stage process comparability work required by regulators. This work highlights the CMC robustness of the ATX002 capsid while still delivering on increased efficacy and delivery benefits."

Engineered AAV2 capsids have improved performance compared to natural serotypes via intra-CSF delivery in both mouse and NHP central nervous system
(ASGCT 2026)
- "Results When packaged, empty/full ratios of ATX001 and ATX002 are significantly improved compared to the natural AAV2 serotype, allowing for production of higher titers for CNS delivery...Furthermore, enhanced packaging of these capsids reduces the cost associated with manufacturing. These findings highlight the strength of Avista’s platform to generate highly manufacturable and potent next-generation AAV vectors for brain-wide gene therapy using lower doses via intra-CSF administration."

Structural dynamics insights into principles underlying the fitness of new broadly potent AAVs.
(PubMed, bioRxiv)
- "ATX002, the top-performing vector, demonstrates broad potency across species, including nonhuman primate, mouse, and human, as well as across retina and brain...Structural dynamics analysis revealed a bifunctional molecular mechanism that confers potency through increased affinity of the capsid to the AAV receptor and regulation of heparan sulfate binding. This work provides critical insights relating structural mechanism to the fitness of engineered AAVs and establishes rich new avenues for AAV engineering through the integration of sequence-level analysis with computational biophysics."
