The Sequence [RRKLPVGRS] Is a Nuclear Localization Signal for Importin 8 Binding (NLS8): A Chemical Biology and Bioinformatics Study.
(PubMed, Int J Mol Sci)
- "The sequence was validated in vitro in the breast cancer cell line T47D. This work subscribes to the effort to clarify the intricate relationships between nuclear transporters and their cargo proteins, in order to better understand the mechanism of nuclear transport of proteins and lay the groundwork for the development of novel therapeutics that target particular importins and have an immediate translational impact."

Therapeutic mitigation of thetoxic phenotype induced by TDP-43in animal models of ALS
(ALS-MND 2023)
- " To test this, we studied (1) the protein-proteininteraction between NF242 and TDP-43in vitroandin silico,(2) the co-expression of RGNEF or NF242 with TDP-43 in Drosophila melanogaster, and (3) the viral ectopic expressionof NF242 in an aggressive murine model of ALS (r NLS8)... We observed that NF242 interacts directly with TDP-43 and that the RNA recognition motifs (RRM) domains of TDP-43 and the IPT/TIG domain of NF242 are essential forthis interaction. In vivoco-expression of NF242 with TDP-43in Drosophila melanogaster suppresses its deleterious pheno-type including increasing lifespan, abolishing motor defects, and preventing neurodegeneration. Intracerebroventricularinjection of AAV9/NF242 in a severe TDP-43 murine model of ALS was associated with improvement in the lifespan andmotor phenotype and a decrease in neuroinflamma-tion markers."

Effects of NLS-8 (Melafenoxate) on Memory in a Model of Alzheimer’s Disease, the Scopolamine-Induced Amnesia in the Novel Object Recognition Test in Mice
(ASCP 2023)
- "Donepezil, one of the most used compound for treatment of dementia due to Alzheimer's Disease (AD), has been shown to improve scopolamine-induced amnesia in rodents. The results of this pre-clinical study show that NLS-8 improved amnesia induced by scopolamine suggesting that NLS-8 may improve memory and reduce cognitive symptoms of AD. These effects were significant at doses of 50 and 150 mg/kg and was close to significant at the dose of 100 mg/kg. Learning Objectives Describe the specific pharmacology of NLS-8 Describe the process by which NLS-8 is developed"
