Immunotherapy for Newly Diagnosed Glioblastoma: Current Evidence and Future Perspectives.

- " Six therapeutic classes were represented: immune checkpoint inhibitors (13 reports), peptide and neoantigen vaccines (12), dendritic cell vaccines (10), adoptive cell therapies (5), oncolytic viral and gene therapies (3), and autologous formalin-fixed tumor vaccines (2)... Immunotherapy in newly diagnosed GBM is feasible and tolerable but has not improved survival, with outcomes still determined more by MGMT promoter status and extent of resection than by treatment assignment. Biomarker-enriched selection, lymphocyte-sparing design and rational combinations are required in future trials."

A randomized, placebo-controlled phase III trial of an autologous, formalin-fixed tumor vaccine for newly diagnosed glioblastoma: trial protocol.

- "Given the growing interest in immunotherapy (IMT), we developed an autologous formalin-fixed tumor vaccine (AFTV) manufactured from the patient's own glioblastoma multiforme (GBM) tissue in paraffin-embedded blocks made from the resected tumor and a double-blind, randomized phase IIB trial of AFTV with temozolomide in newly diagnosed GBM was conducted. The 3-year progression-free survival (PFS) rate for patients with gross total resection (GTR) on imaging tended toward improvement: 81% in the AFTV group versus 46% in the placebo group (P = .067). Based on these IIB results, the feasibility of conducting a phase III trial was confirmed for IIB-eligible patients with total resection. Eradication of breast carcinoma with bone metastasis by autologous formalin-fixed tumor vaccine (AFTV) combined with radiation and chemotherapy

- "...We then treated her with the aromatase inhibitor, anastrozole...We treated her again with second course of AFTV and palliative radiation therapy (36Gy/12fractions/19days) together with letrozole, bisphosphonate, additional 2 shots of nivolmab (40mg/48kg body) in October and November 2015...Alteration of her blood CEA level suggests that the combined treatments eradicated the metastatic breast carcinoma. In conclusion, the present case suggests that AFTV in combination with palliative X-ray irradiation and adjuvant aromatase inhibitors eradicates skeletal metastasis of breast carcinoma with enhancement of a specific anti-tumor immune reactivity."
