Heterologous prime-boost vaccination against H5 avian influenza: Safety and immunogenicity of a MF59-adjuvanted, cell-culture derived H5N6 vaccine.

- "This phase 2 extension study evaluated immunogenicity and safety of MF59-adjuvanted, cell culture-derived H5N6 vaccine (aH5N6c) in adults primed with MF59-adjuvanted, cell culture-derived H5N1 vaccine (aH5N1c) and in unprimed adults...Solicited adverse events were mild or moderate, comparable between groups, and consistent with other MF59-adjuvanted pandemic vaccines; no vaccine-related serious adverse events occurred. Heterologous H5N6 booster vaccination in H5N1-primed adults elicited strong cross-reactive immunity against the priming strain, demonstrating long-lasting immune memory for at least 6 y and supporting heterologous prime-boost strategies for pandemic preparedness against emerging H5 outbreaks."

- "Use of cell-based instead of egg-based vaccine was estimated to additionally prevent 23,364 episodes of acute otitis media, 12,408 asthma exacerbations, 5,932 episodes of acute bronchitis and 1,937 bacterial pneumonia cases, as well as 92,486 antibiotic prescriptions. Conclusions/Learning Points In the paediatric population, use of a more effective cell-based influenza vaccine could confer substantial public health benefits beyond influenza prevention, through clinically meaningful reductions in secondary complications and antibiotic use, supporting improved antibiotic stewardship."

Economic Impact of Pediatric Influenza Vaccination Coverage in the United States: A Modeling Analysis of Direct Medical Costs

- "At the current U.S. influenza vaccination rate in pediatric population (~50%), total estimated influenza-associated medical costs during a high-incidence season amounted to US$251M for outpatient care and US$530M for inpatient care. In a low-incidence season, these costs were US$88M (outpatient) and US$170M (inpatient). Achieving the Healthy People 2030 target vaccination rate (70%) would yield significant economic savings: approximately US$125M during high-incidence seasons and US$79M during low-incidence seasons.Conclusion(s): Improving pediatric influenza vaccination rates to recommended targets represents a highly cost-effective strategy, with the potential to significantly reduce direct medical expenditures, alleviate healthcare system burden, and support national public health goals."

- "The dataset included 60,990 vaccinated and influenza-tested pediatric patients, of whom 7,194 (11.8%) received QIVc and 53,796 (88.2%) received QIVe. QIVc was significantly more effective than QIVe in preventing test-confirmed influenza in the full pediatric population, with an estimated rVE of 19.6% (95% CI: 13.6-25.3%). Consistent rVE results were observed across both pediatric age subgroups and in the outpatient setting (Figure 1)."
