Lipid nanoparticle-mediated CD45 epitope editing in CD34+ cells protects human hematopoiesis from CART45 therapy (ASGCT 2026) - "In addition, the donor-matched CART45 cells selectively eliminated unedited human hematopoietic cells while providing comparable protection for EP- and LNP- edited cells in vitro and in vivo...Conclusion Together, these data establish LNP-mediated base editing as a streamlined and potentially GMP-adaptable strategy enabled by a simple delivery workflow that preserves long-term HSC function comparable to EP. This LNP-based delivery approach represents a clinically scalable alternative for genetic engineering of HSPCs, supporting standardized tech transfer and reproducible multi-center manufacturing, with broad potential applicability across hematologic malignancies and other blood disorders."
