A Dissolving Microneedle Design for Poorly Water-Soluble Drugs for Enhanced Skin Permeation and Transdermal Delivery Fabricated Using 3D Printing.
(PubMed, Micromachines (Basel))
- "These optimized structures served as templates for flurbiprofen (FLU)-loaded dissolving MNs (DMNs) fabricated via a bilayered casting method...Consequently, star-type DMNs achieved the highest cumulative drug permeation (86.9 ± 9.9% in 12 h), outperforming pyramidal (77.8 ± 9.0%) and conical (64.4 ± 10.2%) designs. These findings underscore geometric design as a critical determinant of clinical efficacy, providing a robust framework for precision manufacturing of task-specific MNs for poorly soluble drugs."

Bifunctional Analgesic-Pro-Healing Hydrogel Dressing for Postoperative Analgesia and Wound Healing in Mice.
(PubMed, ACS Biomater Sci Eng)
- "In vitro and in vivo experiments have confirmed that hCMPG-FP can exert dual functions of wound healing promotion and analgesia, which is associated with the antibacterial activity, coagulation-promoting effect, and cell migration-promoting ability of carboxymethyl chitosan hydrogel, as well as the analgesic and anti-inflammatory properties of flurbiprofen. In conclusion, when acting on incisions, hCMPG-FP regulates multiple pathways such as wound healing and inflammation modulation, exerts ideal dressing functions, and provides a theoretical basis and experimental evidence for the further development of innovative wound treatment strategies."

Low-dose dexmedetomidine improves postoperative sleep and pain in gynecological surgery: a randomized trial.
(PubMed, Front Pharmacol)
- P=N/A | "Secondary outcomes included PSQI scores on the first and second postoperative nights, Visual Analogue Scale (VAS) pain scores assessed at 6, 12, 24, and 48 h postoperatively, total postoperative sufentanil consumption, the number of PCA button presses recorded by the infusion pump, and the incidence of adverse events-specifically bradycardia (heart rate <50 bpm), hypotension (mean arterial pressure <60 mmHg), postoperative nausea and vomiting (PONV), and the use of rescue analgesia with intravenous flurbiprofen axetil. Collectively, our findings support that this low-dose regimen is a safe and effective multimodal analgesic strategy, offering a valuable therapeutic option to simultaneously improve sleep and alleviate pain in female patients during the postoperative period. https://www.chictr.org.cn, identifier ChiCTR2500108204."

Repurposing flufenamic acid as a putative PmrB-directed adjuvant to restore colistin activity in Klebsiella pneumoniae.
(PubMed, Microbiol Spectr)
- "High-throughput virtual screening of DrugBank using integrated ligand- and structure-based approaches shortlisted five approved drugs (mebendazole, flurbiprofen, tirbanibulin, flufenamic acid, and netarsudil) for experimental evaluation. Mechanistically, our findings are consistent with modulation of the PmrB/PmrA resistance pathway, supporting bacterial histidine-kinase signaling as a promising direction for antibiotic adjuvant development. Overall, this study highlights a practical and scalable framework for discovering resistance-targeted adjuvants to help protect last-resort antibiotics and improve treatment options for difficult-to-treat infections."

nido-carborane conjugates with ibuprofen, flurbiprofen and fenoprofen: synthesis, characterization, COX inhibition potential and anticancer activity.
(PubMed, J Inorg Biochem)
- "Furthermore, the nido-carborane conjugates with ibuprofen, flurbiprofen or fenoprofen incorporating propylene spacers 3b, 6b, and 9b, respectively, displayed the most potent activity against A549 lung cancer cells and in comparison to other tested molecules exhibited better selectivity towards malignant cells, as evidenced by selectivity index values. The main mechanism of action of these NSAID conjugates includes caspase dependent apoptosis synchronized by inhibited cell division and hyperproduction of superoxide anions, hydrogen peroxide and peroxynitrites."

A predictive release kinetic model guiding the rational design of sustained release solid dispersions with insoluble carriers.
(PubMed, Int J Pharm)
- "Flurbiprofen (FBU) is a nonsteroidal anti-inflammatory drug with short half-life, requiring frequent dosing and leading to potential gastrointestinal side effects...The model was further utilized to predict the release behavior for varying carrier compositions and particle sizes successfully, demonstrating its ability in guiding the rational design of formulations based on insoluble polymeric carriers. This work underscores the critical role of mechanistic modeling in transitioning from empirical formulation screening to rational design."
