- "DIO2 mRNA expression and enzyme activity were measured in primary CAF cultures and in tumors from TBP mice treated with cefuroxime (DIO2 inhibitor) alone or in combination with lenvatinib (multikinase inhibitor). CAF-specific DIO2 expression is observed across many human tumor types and is associated with a pCAF transcriptome profile. Our in vivo and in vitro DIO2-expressing CAF models enable us to study the functional role of CAF DIO2 in tumor progression. Targeting DIO2 may represent a novel strategy for selectively inhibiting or reprogramming pCAFs while preserving tumor-restraining CAF populations, addressing a major challenge in CAF-directed therapy."
