Establishment of a Visible Reporter System in Zymomonas mobilis through Random Mutagenesis and Rational Design of a Chromoprotein eforRed.
(PubMed, ACS Synth Biol)
- "Mechanistic studies revealed that substitutions at the amino acid residues K201 and T24 situated on the surface region of eforRed, especially the double-mutant K201H-T24V, could optimize the protein optical characteristics by concurrently increasing intrinsic fluorescence and elevating protein expression levels. Finally, a visible reporter-gene system was successfully established in Z. mobilis by expressing the mutant eforRedK201H-T24V under a strong promoter like Pgap-4S or PT7 that gave colonies a visible red color for phenotypic screening and retained high fluorescence intensity for quantitative analysis. This study thus not only constructed a robust visible reporter-gene system in the non-model bacterium Z. mobilis but also helped expand the applications of chromoproteins in biotechnology and synthetic biology, especially in non-model microorganisms."

Simultaneous optical recording of action potentials and calcium transients in cardiac single cells differentiated from type 1 CPVT-iPS cells.
(PubMed, Front Physiol)
- "In drug-loading experiments on CPVT1 (RyR2-I4587V) hiPSC-derived ventricular-like CMs, carvedilol and flecainide demonstrated some effectiveness, while JTV519 at 3 µM exhibited both efficacy and alterations in AP waveforms. The Ca2+/calmodulin-dependent serine-threonine protein kinase II (CaMKII) inhibitor KN-93 at 1 µM was highly effective (93%) at reducing Ca2+ transient abnormalities without altering AP waveforms."

Development of novel InDel markers by whole-genome sequence comparison and genetic diversity assessment of Thailand rice blast fungus populations.
(PubMed, Stress Biol)
- "The effectiveness of these 82 loci was assessed by analyzing the genetic diversity of 47 Thai rice blast isolates alongside two reference isolates, GUY11 (France) and KJ201 (Korea)...This finding suggests a correlation between genetic relatedness and the plant hosts susceptible to rice blast disease. The newly developed InDel markers provide a valuable resource for future research in this field."

c-di-GMP inhibits rRNA methylation and impairs ribosome assembly in the presence of kanamycin.
(PubMed, EMBO Rep)
- "Molecular dynamics simulation and mutagenesis studies reveal that c-di-GMP binds to RlmI at residues R64, R103, G114, and K201...Binding and methylation assays indicate that the inhibitory effect of c-di-GMP on RlmI is conserved across various pathogenic bacteria. Our data suggest an unexpected role for c-di-GMP in regulating ribosome assembly under stress through the inhibition of rRNA methyltransferases."

Disruption of Ca2+ Homeostasis and Enhanced Ca2+ Signaling in Scleroderma Fibroblasts Is Due to the Vicious Cycle Formed by Mitochondrial-derived Reactive Oxygen Species, CaMKII, and ER Extrusion Pump RYR2
(ACR Convergence 2024)
- "Fibroblasts were treated with a CaMKII inhibitor KN93, a mitoROS quencher Mitoquinol (MitoQ), or a RYR2 stabilizer JTV519 for 72 hours. Our research revealed a new paradigm where intracellular Ca2+ and CaMKII foster a pro-fibrotic profile in SSc fibroblasts. We provided evidence that the dual activation of CaMKII – via Ca2+/calmodulin binding and oxidative modulation – amplifies Ca2+ signaling, which in turn drives the fibrotic phenotype. These insights significantly extend the current understanding of Ca2+-related mechanisms in SSc fibrosis, a previously underexplored area."

Mitochondrial MOF regulates energy metabolism in heart failure via ATP5B hyperacetylation.
(PubMed, Cell Rep)
- "We demonstrate that the acetylation of ATP5B at K201, co-regulated by MOF and SIRT3, impairs mitochondrial respiration and energy metabolism both in vitro and in vivo. These findings suggest that the role of MOF in HF could be attributed to its regulation of ATP5B acetylation. Overall, our results highlight the disruptive impact of mitochondrial MOF on cardiac function and emphasize the significance of enzyme-catalyzed acetylation in mitochondria."

None of the patients complained of discomfort during the procedure, and in all cases, margins were found to be free from carcinoma as determined by final pathology Results .ConclusionWe recommend this localization technique using mixture of indigo-carmine and lipiodol in concert with the transbronchial approach because the procedure time is short, patient's discomfort is low, and success rate is high."

Improved Antioxidative, Anti-Inflammatory, and Antimelanogenic Effects of Fermented Hydroponic Ginseng with Bacillus Strains.
(PubMed, Antioxidants (Basel))

After fermentation by B. subtilis KU201, the antioxidant activity, determined using ABTS and FRAP assays, increased from 25.30% to 51.34% and from 132.10% to 236.27%, respectively, accompanied by the enhancement of the phenolic compounds and flavonoids...These results indicate that the HG exhibited stronger antioxidative, anti-inflammatory, and antimelanogenic effects after fermentation. Consequently, HG fermented by Bacillus strains can potentially be used as an ingredient in cosmetological and pharmaceutical applications."
