Interleukin 4 Promotes Hypercontractility in Human Airway Smooth Muscle Cells

- " Pathway analysis revealed that IL-4 significantly changed genes related to ion channels/Ca2+ regulation (KCN1, ANO1, ANO3), Rho/Rap GTP signaling (ARHGEF35, RAPGEF4) and cytoskeletal protein regulation (CRACD, ITGA7 and ITGB7)... Our novel data shows that exposure to IL-4 can promote human ASM hypercontractility through increased α-SMA expression and actin polymerization. These pro-contractile effects were abrogated by a RhoA inhibitor, suggesting a common mechanism with TGF-β1. Our study suggests that IL-4 may be a key type 2 pro-inflammatory cytokine in promoting airway hypercontractility in asthma."
