Slowly Burning Down the House: The Role of Calcium in Spotted Fever Rickettsiosis Pathogenesis

- "Additionally, in a dose-dependent manner, both benidipine and nifedipine (L-type VGCCb) delayed R. parkeri-induced VP and bacterial growth...Replicating but not heat-killed R. parkeri caused EC VP; bacterial protein synthesis was necessary for EC barrier dysfunction since subinhibitory doxycycline doses delayed R. parkeri-induced EC dysfunction without inhibiting bacteria growth...parkeri EC infection leads to calcium entry and ATP consumption that promote fitness; limiting compensatory calcium influx by VGCCbs impairs bacterial division, hinders bacterial spread cell-to-cell, disrupts growth, and mitigates R. parkeri-induced EC VP. These findings suggest that rickettsial manipulation of host cell homeostasis and calcium flux is key to rickettsial fitness, pathogenicity, and disease severity."

- "The inhibitory mechanism of mobocertinib was further investigated for nicardipine, irbesartan, telmisartan, carvedilol, prazosin hydrochloride, and spironolactone. In this study, the most common antihypertensive drugs were found to inhibit the metabolism of mobocertinib in vitro. We found that benidipine, nicardipine, telmisartan, and irbesartan inhibited the metabolism of mobocertinib in vitro and in vivo."
