DNA Aptamers with Chemically Locked Ends for Virus Infection Inhibition.

- "Diagonal T-T bases were designed in the ends of aptamers and could be chemically cross-linked with trioxsalen via photocatalyzed cycloaddition...Notably, in the application exploration, the terminal locked aptamers, which bound to receptor-binding domains on SARS-CoV-2, showed superior performance in virus infection inhibition. This work puts forward a paradigm to develop a biological regulation pattern for aptamers based on chemical terminal manipulation of DNA, potentially promoting the clinical applications of nucleic acid drugs."

- "Indeed, the few tools available currently could only investigate unwound (Trioxsalen) or overwound (GapR) DNA topology (variations in twist) but not the variations in writhe (supercoils and plectonemes)...TRF2 depletion caused both telomere relaxation and an increase in positive supercoils while the inhibition of Histone Deacetylase I and II by TSA only caused telomere relaxation. Moving outside telomeres, we also observed a build-up of positive supercoils on the FRA3B fragile site following replication stress, suggesting a topological model of DNA fragility for this site."

- "Psoralens and their derivatives, such as trioxsalen, have unique crosslinking features to DNA. We found that the introduction of an oligonucleotide via a linker at the C-5 position of psoralen can promote favorable crosslinking to target double-stranded DNA. We believe our findings are essential information for the development of Ps-Oligos as novel gene regulation tools."

Design and Application of Mini-libraries of the miRNA Probes for an Efficient and Versatile miRNA-mRNA Cross-linking.

- "The new strategy is based on the utilization of a controlled mixture of probes for a chosen miRNA, in which the trioxsalen moiety is introduced at the N 4 -position of a selected cytidine through short oligoethylene glycol-based linkers. In vitro photo-cross-linking experiments with mini-libraries of probes for microRNAs of interest showed variable cross-linking efficiencies, giving strong evidence for the general applicability of the presented approach."
