Therapeutic Considerations of Sleep Disturbances in Depression
(WFSBP 2025)
- "Concerning, pharmacological approaches, there has been a focus on low-dose sedative antidepressants like trazodone, doxepine, mirtazapine or trimipramine. It is strongly recommended to treat insomnia in MDD, specifically, in order to relieve symptoms of depression and to foster remission. CBT-I or medication with low dose sedative antidepressants are effective and well tolerated, while chronic treatment with GABA-receptor-agonists is not recommended because of development of tolerance and withdrawal insomnia."

Repurposing Tricyclic Antidepressants for Glioblastoma Treatment
(EACR 2025)
- "Our lab has previously proposed a successful novel combinatorial regimen, including the tricyclic antidepressant (TCA) imipramine, the vascular endothelial growth factor (VEGF) inhibitor bevacizumab, and the immune checkpoint inhibitor anti-PD-L1, which demonstrated significant therapeutic benefit in GBM mouse models (Chryplewicz et al.; Cancer Cell, 2022)...Motivated by these results and encouraged by the clinical support, we aim to further improve this therapeutic regimen by disrupting various capabilities of tumors required for their growth and therapeutic resistance.Material and To optimize this regimen for clinical translation, we screened multiple TCAs - trimipramine, clomipramine, and amitriptyline - alongside imipramine in mouse and human GBM cell lines to evaluate their impact on cell viability... This study aims to delve deeper into TCA mechanisms of action, assess potential synergies with VEGF pathway inhibitors and immune checkpoint blockade, and ultimately..."

Pharmacotherapy of psychiatric inpatients with mental and behavioral disorders caused by sedatives or hypnotics (F13): Results from an observational pharmacovigilance program between 2000 and 2017.
(PubMed, Addict Sci Clin Pract)
- "We found high rates of non-SHA drugs among patients treated for SHA-SUD. The prevalent use of psychotropic drugs with strong sedating properties indicates a symptom-oriented treatment approach, which is often "off-label" but may be clinically necessary. Main limitation is the cross-sectional design of the AMSP databank. Therefore, we cannot provide any follow up data on the patient collective especially regarding outcome."

The cardiovascular safety of tricyclic antidepressants in overdose and in clinical use.
(PubMed, Ther Adv Psychopharmacol)
- "Amitriptyline, clomipramine, dothiepin/dosulepin, doxepin, trimipramine and imipramine showed broadly similar toxicity and were usually reported to be less toxic than desipramine...With respect to the risk of cardiovascular events at clinically relevant exposure, amitriptyline, nortriptyline and lofepramine were associated with a greater risk of in-use cardiotoxicity. All measures of overdose toxicity were subject to external influences and confounding. The continued use of TCAs in depression and other conditions should be minimized when considering their undoubted toxicity in overdose and possible toxicity in normal clinical use."

Development of Visible Spectrophotometric Methods for the Determination of Tricyclic Antidepressants Based on Formation of Molecular Complexes with p-Benzoquinones.
(PubMed, Int J Mol Sci)
- "The present work describes two visible (VIS) spectrophotometric techniques that utilize the formation of charge transfer complexes between four antidepressant compounds, namely, amitriptyline hydrochloride (AMI), imipramine hydrochloride (IMI), clomipramine hydrochloride (CLO), and trimipramine maleate (TRI) acting as electron donors and two p-benzoquinones, namely, p-chloranilic acid (pCA) and 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), serving as electron acceptors. The developed spectrophotometric techniques were validated intra-laboratory and successfully applied for quantitative assessment of the four antidepressant active ingredients in several commercial pharmaceutical formulations. The methods are relatively simple, fast, and use readily available laboratory instrumentation, making them easily applicable by most quality control laboratories worldwide."

Potential of sodium dodecyl sulfate micellar solutions as eluents in magnetic dispersive micro-solid phase extraction with polydopamine-coated magnetite nanoparticles. Application to antidepressant drugs.
(PubMed, J Chromatogr A)
- "In this paper, the potential of micellar solutions of the anionic surfactant sodium dodecyl sulfate (SDS) as eluents in dispersive micro-solid phase extraction (D-μSPE) using polydopamine-coated magnetite nanoparticles (FeO@PDA NPs) for the extraction and preconcentration of seven basic drugs (bupropion, citalopram, fluoxetine, mianserin, nomifensine, trimipramine, and viloxazine) is explored for the first time (to the best to our knowledge) and compared with conventional hydro-organic eluents. The results shown in this paper demonstrate the suitability of FeO@PDA NPs as a sorbent for the extraction of antidepressants as well as the advantages of using SDS micellar solutions over classic hydro-organic eluents containing methanol, acetonitrile or tetrahydrofuran. Finally, the stability and reusability of the FeO@PDA NPs is proven."

Drug triggered pruritus, rash, papules, and blisters - is AGEP a clash of an altered sphingolipid-metabolism and lysosomotropism of drugs accumulating in the skin?
(PubMed, Lipids Health Dis)
- "Amiodarone, chlorpromazine, amitriptyline, and trimipramine are classified lysosomotropic as well as photosensitizing, however, they fail to trigger rash and pruritic papules in all individuals. The suggested model of a compartmentalized ceramide metabolism provides a more sophisticated explanation of cutaneous drug adverse effects and the individual sensitivity to UV radiation.
