Investigation of the Putative Relationship Between Copper Transport and the Anticancer Activity of Cisplatin in Ductal Pancreatic Adenocarcinoma.

- "Two major strategies were pursued: (i) inhibiting copper transporters ATP7A and B with tranilast (TR) and omeprazole (OM) to block the cellular copper and, potentially, also cisplatin efflux, and (ii) using the chelator elesclomol (ES) to elevate intracellular copper and cisplatin levels. Moreover, all drugs contributed to the enhanced cellular anticancer activity of cisplatin, with ES being the most effective compound. The results suggest that the targeted modulation of copper transport mechanisms may offer novel adjuvant approaches for the treatment of PDAC."

- "This study aimed to explore whether TRPV2 contributes to SU5416/hypoxia (SuHx)-induced ventricular remodeling and arrhythmias, and its possible mechanisms...In the SuHx-induced PH rat model, TRPV2 inhibitor tranilast significantly reduced mean pulmonary artery pressure (mPAP) and pulmonary vascular remodeling; relieved ventricular remodeling, myocardial fibrosis, and hypertrophy; shortened the prolongation of QT, QTc, Tpeak-Tend interval, effective refractory period (ERP) and action potential duration (APD); reduced susceptibility to ventricular fibrillation (VF)...TRPV2 inhibition suppressed the IGF1-mediated Wnt/β-catenin signaling pathway. Our data support TRPV2 inhibition prevents PH-induced right ventricular remodeling and VF susceptibility; TRPV2 can serve as a therapeutic target for PH-induced VF."

- "The inhibition of either TRPV2 with tranilast or a disintegrin and metalloprotease domain-containing protein 10 (ADAM10) with GI254023X significantly reduced H2O2-induced VE-cadherin cleavage, while TRPM2 inhibition by econazole significantly increased H2O2-driven VE-cadherin cleavage and blockage of TRPV4 showed no effect...This process of cadherin-switching did not occur upon inhibition of TRPV2 or ADAM10. Our results highlight a novel role for TRPV2 as a redox sensitive ion channels in the microvascular endothelium and provide insight into the mechanisms underlying pulmonary microvascular endothelial barrier recovery."

Dual inhibition of TGFβ and PDGF improves RV remodeling and function in response to RV pressure or volume-loading.

- "Here, we investigate whether dual inhibition of TGFβ and PDGF, using Tranilast (TRN), improves RV remodeling in response to pulmonary artery banding (PAB) or pulmonary regurgitation (PR)...We identify that PDGF drives RV fibroblast proliferation and activation via SMAD2/3, JNK, and β-catenin signaling. Our studies suggest that TGFβ and PDGF are interconnected drivers of RV fibrosis and hence synergistic targets to improve RV remodeling in RV pressure and volume loading."
