- "The HIV vaccine field has swung between humoral-based to cell-mediated approaches; however the consensus has evolved toward the understanding that an induction of both may be important for optimal protection...Notably, all A*01:01 T cell epitopes overlapping bNAb epitopes were within conserved ENV regions, and all but one involved glycan-dependent bNAb binding. The epitope landscape analysis therefore showed the allele associated with slower HIV-1 infection rates presents a narrower repertoire of ENV CD8+ T cell epitopes, that preferentially overlaps bNAb target sites, consistent with coordinated immune targeting between cellular and humoral arms of immunity."

- "While therapeutic vaccines are a promising strategy for inducing human immunodeficiency virus (HIV) control, HIV vaccines tested to date have offered limited benefit to people living with HIV...Combination with FLT3Ra also expanded polyfunctional CD4+ T cell responses. Our data demonstrate enhanced and distinct shaping of vaccine-elicited immune responses by immune modulators with implications for developing a functional HIV cure."

[VIRTUAL] A PLACEBO-CONTROLLED ATI TRIAL OF HTI VACCINES IN EARLY TREATED HIV INFECTION

- P1, P2a | "Background: HTI is a novel HIV vaccine immunogen designed at redirecting cellular immune responses to HIV targets associated with viral control. The AELIX-002 trial (NCT03204617) was a randomized, singlecenter, placebo-controlled trial to evaluate the safety, immunogenicity and antiviral effect of DNA.HTI (D), MVA.HTI (M) and ChAdOx1.HTI (C) vaccines after discontinuation of ART in early-treated people living with HIV (PLWH)... HTI vaccines were safe and highly immunogenic in early-treated PLWH with a prolonged time off ART seen in vaccinees with non-beneficial HLA class I alleles. Time off ART positively correlated with vaccine-induced HTIspecific T cell responses at ATI start. Multivariate analysis for other correlates of response is ongoing."
