Bioactive specialized metabolites of Trochila sp., an endophytic fungus of Lilium carniolicum.
(PubMed, Phytochemistry)
- "BGP15P7IS3, an undescribed endophytic fungus from Lilium carniolicum, resulted in isolation and identification of eleven specialized metabolites, of which four pyridone alkaloid derivatives (1, 2, 4, and 5) and a biphenyl derivative (8) were identified as previously undescribed natural products...Compound 3 displayed antifungal activity only against C. neoformans with an MIC value of 12.5 μg/mL, and compound 6 exhibited promising antifungal activity with MIC values of 0.39-3.25 μg/mL against all the aforementioned fungal pathogens, and a concentration-dependent growth inhibitory property against T. vaginalis. Based on the identification of derivatives, a revised biosynthetic pathway for pyridone-type alkaloids was proposed."

(PubMed, Aging Dis)
- "Next, we utilized an OPA1 activator BGP-15, an OPA1 inhibitor myls22 and an OPA1 overexpression virus to investigate the role of OPA1 in AD...This may relate to down-regulation of hexokinase-2 (HK2) expression. BGP-15 improves quality of goat sperm by mitigating oxidative stress during cryopreservation.
(PubMed, Cryobiology)
- "Moreover, the apoptosis rate of freeze-thawed spermatozoa significantly decreased in the 100 μM BGP-15 treatment group compared to the control (P < 0.05), while the MMP of freeze-thawed spermatozoa significantly enhanced in the 100 μM BGP-15 treatment group (P < 0.05). In conclusion, BGP-15 enhances cryo-protective effects on goat spermatozoa, and 100 μM BGP-15 addition to the extender during cryopreservation is beneficial to the goat breeding industry."

The Impact of YabG Mutations on Clostridioides difficile Spore Germination and Processing of Spore Substrates.
(PubMed, Mol Microbiol)
- "Spores derived from C. difficile yabGC207A (a catalytically inactive allele), C. difficile yabGA46D, C. difficile yabGG37E, and C. difficile yabGP153L strains germinated in response to taurocholic acid alone...Finally, changes in yabG expression under the mutant promoters were analyzed using a SNAP-tag and revealed expression differences at early and late stages of sporulation. Overall, our results support and expand upon the hypothesis that YabG is important for germination and spore assembly and, upon mutation of the processing site, can shift where it cleaves substrates."

REPRODUCTIVE AGEING: BGP-15 mitigates adverse impacts of aging on sperm quality, fertility, and offspring health in male mice.
(PubMed, Reproduction)
- "BGP-15 treatment for 3 weeks prior to conception improved on-time pre-implantation embryo development and fetal viability and increased fetal size in pregnancies sired by old males. These results validate that ageing negatively affects male fertility and offspring physiology and indicates that pre-conception treatment with BGP-15 has the potential to improve sperm quality as well as early embryo development and post-natal health."

CAS3D: 3D quantitative morphometry on Second Harmonic Generation image volumes from single skeletal muscle fibers.
(PubMed, Comput Biol Med)
- "The software was applied to a previously published data set of single skeletal muscle fiber volumetric SHG image data from a rat intensive care unit (ICU) model of ventilator-induced diaphragm dysfunction (VIDD) with treatment regimens involving the small anti-inflammatory molecules BGP-15, vamorolone, or prednisolone. Our method reliably reproduced the results of the previous work and improved the standard deviation of the cosine angle sum detection in all sample groups from a mean of 0.03 to 0.008. This improvement is achieved by applying analysis algorithms to the whole volumetric images in 3D in contrast to the previously common method of slice-wise XY analysis."

Altered Autophagic Flux in GNE mutant cells of Indian Origin: potential drug target for GNE Myopathy.
(PubMed, Exp Cell Res)
- "Interestingly, treatment of GNE mutant cells with HSP70 activator, BGP-15, restored the expression and refolding activity of HSP70 along with autophagosome formation. Treatment with calcium chelator, BAPTA-AM restored the cytoplasmic calcium levels and autophagosome formation but not LC3II/I ratio significantly. Our study provides insights towards GNE mutation specific response for autophagy regulation and opens up a therapeutic advancement area in calcium signalling and HSP70 function for GNE related Myopathy."

Potential small effector molecules restoring cellular defects due to sialic acid biosynthetic enzyme deficiency: Pathological relevance to GNE myopathy.
(PubMed, Biochem Pharmacol)
- "In the present study, we analyzed the effect of small molecules like BGP-15 (HSP70 modulator), IGF-1 (IGF-1R ligand) and CGA (cofilin activator) on cellular phenotypes of GNE heterozygous knock out L6 rat skeletal muscle cell line (SKM‑GNEHz)...Treatment with CGA recovered epimerase activity by 49 %, F-actin assembly by 132 % and cell migration upto 41 % (24 h) in SKM‑GNEHz. Our study shows that treatment with these small effector molecules reduces the detrimental phenotype observed in SKM‑GNEHz, thereby, providing insights into potential therapeutic targets for GNEM."
