Analysis of Laparoscopic and Transvaginal Ovarian Drilling on Polycystic Ovarian Syndrome: A Narrative Review of Outcomes.
(PubMed, Cureus)
- "Laparoscopic ovarian drilling (LOD) and transvaginal ovarian drilling (TVOD) are widely used second-line treatments for ovulation induction in PCOS patients who do not respond to clomiphene citrate (CC)...LOD may be more suitable for patients with elevated luteinizing hormone (LH) levels and shorter durations of infertility. In contrast, TVOD may be preferred for patients who would benefit from a less invasive approach with a lower risk of adhesion formation."

Ursolic acid, a pentacyclic triterpene from Ochrosia elliptica Labill leaves and its role in the management of polycystic ovary syndrome (PCOS).
(PubMed, Inflammopharmacology)
- "Using adult female Wistar Albino rats, PCOS was induced through letrozole administration. The rats were then treated with UA at two different doses (25 and 50 mg/kg), alongside a control group and a standard ovulation-inducing medication, clomiphene citrate (1 mg/kg)...Furthermore, UA downregulated key genes involved in steroidogenesis and oxidative stress response, suggesting a multifaceted mechanism of action. The findings highlight UA's potential as a novel therapeutic option for managing PCOS symptoms, emphasizing the need for further research into its efficacy and safety in clinical applications."

From tablets to targeted therapy: the APHRODITE era in male infertility.
(PubMed, Hum Reprod)
- "While clomiphene citrate and aromatase inhibitors retain a role in specific endocrine profiles, the new positioning of exogenous gonadotropins-LH/hCG and FSH-as targeted, earlier-line interventions holds promise to improve outcomes and efficacy in male fertility care. Future progress will depend on prospective trials that apply the APHRODITE criteria and on comparative analyses of oral agents versus injectable gonadotropin regimens in well-defined patient phenotypes."

Cancer Incidence in Women After Medically Assisted Reproduction.
(PubMed, JAMA Netw Open)
- "Three MAR cohorts were created: assisted reproductive therapy (ART), intrauterine insemination with ovarian stimulation (IUI/OS), and ovulation induction using clomiphene citrate (clomiphene citrate)...The incidence of certain cancers appeared elevated; however, the excess numbers of these cancers were small, and there was reduced incidence of other cancers. Causation cannot be inferred from this descriptive evidence, but findings may guide women and their health care practitioners."

Luteal Phase Support in Ovulation Induction Cycles: Current Evidence and Controversies
(ISGE 2026)
- "While gonadotropin-stimulated cycles are known to cause luteal phase insufficiency, the need for progesterone supplementation in cycles using oral agents such as clomiphene citrate or letrozole is less clear. Luteal phase support is beneficial and recommended for gonadotropin-stimulated OI-IUI cycles. However, its routine use in clomiphene or letrozole cycles is not supported by current evidence. Further well-designed randomized studies are needed to clarify the role of LPS in aromatase inhibitor–based stimulation and to determine the optimal dose and duration across different stimulation protocols Keywords: luteal phase support, ovulation induction, progesterone, clomiphene, letrozole, intrauterine insemination"

Polycystic Ovarian Syndrome - exploring the therapeutic and biomedical insights into the role of Silver nanoparticles.
(PubMed, J Steroid Biochem Mol Biol)
- "Conventional medicines, such as metformin, clomiphene citrate, letrozole, flutamide, and rosiglitazone, can regulate menstrual irregularities, improve ovulation, reduce insulin resistance, and decrease androgen levels, but may also cause various side effects. The review includes evidence from insulin, inflammation, oxidative processes, neuroendocrine factors and cystogenic pathways to combine and provide an overview. The review also discusses how to approach toxicity, especially dose-dependent, and the reproductive safety of AgNPs and their translational limitations to define the boundaries of AgNPs in the treatment of PCOS."

It was concluded that both the regimes i.e. combination of CC plus Letrozole and Letrozole-alone were equally efficacious and safe to use in the treatment of female infertility with Letrozole-alone regime being slightly more effective in terms of endometrial thickness at trigger."

Significance of Follicle-Stimulating Hormone Receptor Gene Single-Nucleotide Polymorphism rs6165/rs6166 Analysis for Infertility-Associated Ovarian Disease Susceptibility Prediction and Optimized Individualized Ovulation Induction/Ovarian Stimulation.
(PubMed, Diagnostics (Basel))
- "Recent research clarifies the association of rs6165/rs6166 with susceptibility to infertility-associated ovarian diseases, ranging from polycystic ovarian syndrome, premature ovarian insufficiency, endometriosis, to ovarian cancer, along with response/resistance to ovulation induction/ovarian stimulation with clomiphene citrate, letrozole, metformin, FSH preparations, and adjunctive growth hormone in infertility treatment. The re6165/rs6166 genotype-guided choice of individualized ovulation stimulation preparations has great potential to reduce unexpected poor or high ovarian responses in ovulation induction and ovarian stimulation and improve clinical outcomes in reproductive medicine. Current evidence is insufficient, and further studies are warranted to ascertain its potential for clinical implementation."

Ameliorative effects of Corchorus olitorius leaf ethanolic extract on letrozole-induced polycystic ovary syndrome (PCOS) in rats.
(PubMed, Avicenna J Phytomed)
- "36 female Wistar rats were divided into six groups (n=6) including Sham, PCOS + vehicle (Dimethyl sulfoxide), PCOS + Clomiphene citrate, PCOS + 200, 400, and 600 mg/kg Corchorus olitorius. olitorius leaf extract showed its ameliorative effects in terms of normalizing the altered vaginal cytology, restoring most parameters, and improving the appearance of ovarian and uterine morphology. These results suggest that C. olitorius leaves ameliorate PCOS symptoms in the studied biochemical and histological parameters."
