A p38 MAP kinase inhibitor suppresses osteoclastogenesis and alleviates ovariectomy-induced bone loss through the inhibition of bone turnover.

- "Furthermore, BMS-582949 administration prevents bone loss in ovariectomized mouse mode by inhibiting both bone resorption and bone formation in vivo. Taken together, these findings indicate that BMS-582949 may be a potential and effective drug for the therapy of osteolytic diseases."

ASK1 inhibitor NQDI‑1 decreases oxidative stress and neuroapoptosis via the ASK1/p38 and JNK signaling pathway in early brain injury after subarachnoid hemorrhage in rats.

- "The use of the p38 inhibitor BMS‑582949 or the JNK inhibitor SP600125 led to significant decreases in the protein expression levels of p‑p38 or p‑JNK, respectively, and a significant reduction in oxidative stress and neuronal apoptosis; however, these inhibitors did not demonstrate an effect on p‑ASK1 or ASK1 protein expression levels. In conclusion, treatment with NQDI‑1 improved neurological function and decreased oxidative stress and neuronal apoptosis in EBI following SAH in rats, possibly via inhibition of ASK1 phosphorylation and the ASK1/p38 and JNK signaling pathway. NQDI‑1 may be considered a potential agent for the treatment of patients with SAH."

[VIRTUAL] Syndecan-1 Promotes Epithelial Cell Senescence Through p38MAPK Activation During Lung Fibrosis

- " The transcriptomic changes of alveolar type 2 cells (AT2) of bleomycin-injured WT and syndecan-1 deficient (Sdc1-/-) mouse lungs were assessed using single cell RNA sequencing (scRNAseq)...A p38MAPK inhibitor (BMS-582949) was used to confirm the involvement of the p38MAPK signaling pathway... The results suggest that overexpression of syndecan-1 by epithelial cells promotes lung fibrosis through an increase of epithelial cell senescence mediating through the p38MAPK signaling pathway. Our study highlights the functional role of syndecan-1 in lung fibroproliferation. Future works include identifying the specific structure of syndecan-1, or the ectodomain's shedding, responsible for regulating cellular senescence."

Glycyrrhizin administration ameliorates Streptococcus aureus-induced acute lung injury.

- "Furthermore, we used different inhibitors to treat infected-A549 cells and found that BMS-582949 (a p38 inhibitor) is the most effective inhibitor for inhibiting pro-inflammatory cytokines (IL-6, TNF-α and IL-1β) production, which suggests that p38 signaling pathway might be the main pathway for S. aureus-induced inflammation. Collectively, the data demonstrates that GL could mitigate inflammation after S. aureus infection."
