Discovery of the First-in-Class Oral Cyclic Peptide Targeting IL-1β for Hidradenitis Suppurativa Treatment

- "Cellular inhibition : Significant inhibition of IL-1β-induced IL-6 secretion in MRC-5 cells at very low concentrations (IC50 ≈ 1 pM, 5-20 folds more potent than canakinumab) Potent inhibition of IL-1β-induced IL-6 secretion in PBMCs (IC50 < 50 pM, lower than the canakinumab) Metabolic stability : The lead compounds showed excellent metabolic stability in plasma and simulated intestinal fluid with no significant degradation observed over 24 hours...It demonstrates sufficient in vivo half-life and oral exposure, with anti-inflammatory activity observed at low doses in in vivo PD and efficacy models. Given the dominant role of IL-1β in hidradenitis suppurativa pathogenesis, the se lead compounds represent a promising new oral therapeutic option for HS treatment."
