ifabotuzumab ADC / Humanigen - LARVOL DELTA

PET/MRI imaging in an intracranial tumour model to assess uptake of 89 Zr-Df-ifabotuzumabpyrrolobenzodiazepine across the blood brain barrier (EANM 2026) - "3T MRI (nanoScan, Mediso, Hungary) was used to measure tumour growth weekly using Dotarem contrast agent. An orthotopic human GBM tumour model in BALB/c nude mice showed comparable tumour uptake between 89Zr-Df-ifabotuzumab and 89Zr-Dfifabotuzumab-PBD. This model offers a great opportunity to study the conditions that allow ifabotuzumab-ADCs and other payloads (225Ac-ifabotumumab) to cross the blood brain barrier. Future studies will include: PET/MRI with a 89Zr-labelled IgG control antibody to assess specificity, tumour uptake in 20-40 mm3 subcutaneous U251 xenografts, and investigating an EphA3- murine GBM model (GL261) to examine orthotopic TME uptake."
