ADCplex K5: A dual-antibody, triple-payload liposomal antibody–drug complex designed to overcome tumor heterogeneity through multi-mechanistic cytotoxic targeting with reduced systemic toxicity
(ESMO 2026)
- "Rather than relying on tightly defined synthetic lethality, the design aims to suppress resistant subclones through concurrent multi-mechanistic cytotoxic pressure delivered at substantially reduced drug exposures. Methods ADCplex K5 incorporates two distinct targeting antibodies together with three cytotoxic payloads: doxorubicin, irinotecan, and monomethyl auristatin E (MMAE). K5-treated cohorts maintained robust myeloid and lymphocyte counts, for preserving immune functions addressing residual disease. Conclusions K5 was rationally designed according to principles established in successful multidrug chemotherapy regimens: complementary mechanisms of action, broad tumor coverage, and minimized overlapping toxicities. This strategy may provide a promising approach for overcoming tumor heterogeneity and resistance while preserving immune function for durable anticancer control."
