Understanding the effects of polymer architecture on antisense oligonucleotide complexation

- "In this talk, I will examine the complexation behavior of a short 16 base pair single stranded antisense oligonucleotide (ASO) payload (EZN-4176, an ASO clinical candidate for modulating castration resistant prostate cancer) with four polymer derivatives with varying polymer architectures (linear homopolymers, linear diblock polymers, and micelles (self-assembled from diblock and triblock polymers))...The effects of the addition order of polymer or micelles with ASOs, as well as the molar ratio of amines to phosphates (N/P ratio), were investigated and analyzed by the formation of polyplexes and micelleplexes respectively. Overall, this work illuminates the preference in the size, shape, and colloidal stability of polyelectrolyte complexes formed with ASOs."
