Examining antifungal activity of elexacaftor/tezacaftor/ivacaftor against Aspergillus fumigatus biofilms

- "We also treated A. fumigatus biofilms with ETA in combination with caspofungin, micafungin, or anidulafungin and assessed growth recovery after treatment...Af106-6.10 is also as or more susceptible to ETI in the CF-relevant stress conditions than standard laboratory conditions... ETI is effective at reducing A. fumigatus biofilm biomass in CF lung-relevant conditions. It increases the efficacy of echinocandins against A. fumigatus biofilms. A CF aspergillosis in vivo mouse model is in early development in our laboratory to test the impact of ETI against A. fumigatus biofilms alone and in combination with echinocandins in the mammalian lung."

The latter pool still cleaves TG substrate, is temporarily protected against antithrombin, and is required for fibrin fiber extension and blood clotting and clot retraction."

- "Collectivity, these data point to the existence of two major pools of proteolytically active thrombin, both of which contributing to thrombin generation, that are formed upon plasma and blood coagulation. One pool of soluble thrombin steers the proteolytic coagulation cascade, while another pool is bound to growing fibrin fibers. The latter pool still cleaves the conventional AMC thrombin substrate, is temporarily protected against antithrombin, and is required for fibrin fiber extension and blood clotting and clot retraction."

- "The frequencies of positive tests were as follows: DP 239/392 (61%), DF 235/389 (60.4%), GC 186/392 (47.4%), AF 106/392 (27%), DD 54/215 (25.1%), BT 16/80 (20%), GM 71/371 (19.1%), AA 10/82 (12.2%) and BG 23/242 (9.5%)... Indoor allergens comprising dust mites (DP and DT), arthropod (GC) and fungi (AF) produced most aeroallergen sensitization in Lagos. These should be prioritized in testing panels in this locality. GM should be selected over BG as outdoor allergen."
