Discovery of putative inhibitors of PBP4 and PBP2a to combat methicillin-resistant Staphylococcus aureus infection | Poster Board #1748 (ACS-Fall 2026) - Aug 24, 2026 - Abstract #4535892; Pres time: Aug 24, 2026; 09:00 PM - 11:00 PM; Location: Hall F2 - POSTERS (McCormick Place Convention Center); "Previously identified scaffold NDM-684 lowers the minimum inhibitory concentration (MIC) of both oxacillin and ceftobiprole (BPR) 16-fold at 60 μM and 25 μM, respectively, and was therefore used as a starting point for a structure-activity relationship study. A library of analogs was synthesized and biologically tested for potentiation of oxacillin and BPR against clinically relevant MRSA isolates, and binding of lead compounds to PBPs was also assessed. Further development of analogs based upon this scaffold that reduce oxacillin and/or BPR MICs below their clinical breakpoints are currently ongoing and are expected to enable potent combination therapies against MRSA."
