CR20BA5-2 effectively and simultaneously blocks IL-23 and TL1A, with each functional arm exhibiting activity comparable to that of the corresponding parental monoclonal antibodies. It demonstrates synergistic or additive therapeutic effects in both in vitro and in vivo experiments, highlighting its potential to improve clinical outcomes in patients with IBD. CR20BA5-2, a 2+2 format bispecific antibody, exhibited comparable potency to monoclonal antibody CR20-M0055hz2 in suppressing the TL1A/DR3-NFκB signaling cascade, TF-1 cell apoptosis and IFNγproduction (Figures A-C)."
