JAK inhibition was associated with clinically meaningful response and complete-response rates in GCA, with manageable safety signals and evidence of glucocorticoid sparing. The strongest evidence supports upadacitinib 15 mg, driven by SELECT-GCA, while smaller real-world cohorts suggest broader class activity in relapsing or refractory GCA. Pooled ORR estimates should be interpreted cautiously because they derive from small observational cohorts, whereas complete-response estimates are strongly influenced by the phase 3 upadacitinib dataset."
